Abstract
An amino acid sequence is described. The amino acid may comprise a signal sequence that is SEQ ID NO. 1a (or a variant or homologue or derivative or fragment thereof) that is expressed as a fusion protein that comprises a protein of interest. The signal sequence directs secretion of the protein of interest. The protein of interest may be a heterologous protein. Secretion of the fusion protein aids purification.
Claims31
1-52. (canceled)
A non-naturally occurring polypeptide having an amino acid sequence comprising SEQ ID No. 5;
- wherein said amino acid sequence is not SEQ ID No. 1 and wherein said amino acid sequence comprises an amino acid sequence that has at least 90% identity with SEQ ID No. 6.
A polypeptide comprising at its N terminal end SEQ ID No. 6;
- wherein said polypeptide is not SEQ ID No. 1.
A polypeptide comprising a signal sequence that causes secretion from both a prokaryotic host and a eukaryotic cell of a protein of interest (POI) when directly fused or indirectly fused to said polypeptide, wherein said polypeptide has an amino acid sequence that is not SEQ ID No. 1.
A method of selecting the expression host for production of a non-naturally occurring polypeptide having an amino acid sequence comprising SEQ ID No. 6;
- wherein said amino acid sequence is not SEQ ID No. 1 and wherein said amino acid sequence comprises an amino acid sequence that has at least 90% identity with SEQ ID No. 6;
- the method comprising the steps of
- i) transforming at least one prokaryotic expression host with a nucleic acid molecule encoding the polypeptide or a vector containing the nucleic acid molecule;
- ii) cultivating the transformed cells to obtain a prokaryotic expression product;
- iii) determining at least one parameter relating to said prokaryotic expression product;
- iv) transforming at least one eukaryotic expression host with a nucleic acid molecule encoding the polypeptide or a vector containing the nucleic acid molecule;
- v) cultivating the transformed cells to obtain the eukaryotic expression product;
- vi) determining at least one parameter relating to said eukaryotic expression product;
- vii) comparing the parameter for the prokaryotic expression product with the parameter for the eukaryotic expression product;
- and
- viii) selecting the expression host based on the comparison.
