Abstract
The present disclosure provides RNA molecules, for example, meroduplex ribonucleic acid molecules (mdRNA), capable of decreasing or silencing BIRC5 gene expression. An mdRNA of this disclosure comprises at least three strands that combine to form at least two non-overlapping double-stranded regions separated by a nick or gap wherein one strand is complementary to a BIRC5 mRNA. Also provided are methods of decreasing expression of a BIRC5 gene in a cell or in a subject to treat a BIRC5-related disease.
Claims24
An RNA molecule that down regulates the expression of a baculoviral IAP repeat-containing protein 5 gene (BIRC5) mRNA, the RNA molecules comprising a first strand that is complementary to a human BIRC5 mRNA as set forth in SEQ ID NO:
- 1158, 1159 or 1160, and a second strand complementary to the first strand, wherein the first strand and second strands can anneal to form a double-stranded region having from about 15 base pairs to about 40 base pairs.
10-11. (canceled)
A meroduplex ribonucleic acid (mdRNA) molecule that down regulates the expression of a baculoviral IAP repeat-containing protein 5 gene (BIRC5) mRNA, the mdRNA molecule comprising a first strand of 15 to 40 nucleotides in length that is complementary to a survivin (BIRC5) mRNA as set forth in SED ID NO:
- 1158, 1159 or 1160, and a second strand and a third strand that are each complementary to non-overlapping regions of the first strand, wherein the second strand and third strand can anneal with the first strand to form at least two double-stranded regions spaced apart by a nick or a gap.
15-16. (canceled)
The mdRNA molecule of clan 17 wherein the at least one acyclic nucleomonomer is selected from the group consisting. of:
- wherein,
- R is selected from the group consisting of hydrogen, methyl group, C(I-IO) alkyl, cholesterol, naturally or non-naturally occurring amino acid, sugar, vitamin, flurophore, polyamine and fatty acid.
(canceled)
A method for reducing the expression of a human BIRC5 gene, comprising administering an RNA molecule to a cell expressing a BIRC5 gene, wherein the RNA molecule comprises at least a first strand and a second strand, the first strand is complementary to a human BIRC5 mRNA as set forth in SEQ ID NO:
- 1158, 1159 or 1160, the second strand is complementary to the first strand, and the first strand and second strands can anneal to form about 15 base pairs to about 40 base pairs, wherein the mdRNA molecule reduces expression of the BIRC5 gene in the cell.
