Abstract
Methods and materials are provided for the production of compositions of erythropoietin protein, wherein said compositions comprise a pre-selected N-linked glycosylation pattern as the predominant N-glycoform.

A method of increasing hematocrit in a mammal comprising administering a therapeutically effective dose of a composition of erythropoietin protein, said composition comprising a plurality of bi-antennary N-linked glycans attached to the erythropoietin, said plurality of N-linked glycans comprising greater than 25 mole percent of an N-linked glycan having the structure of N-acetylglucosamine2Mannose3N-acetylglucosamine2Galactose2N-acetylneuraminic acid, (GlcNAc2Man3GlcNAc7Gal2NANA2), wherein a polyethylene glycol moiety is linked to the N-terminal amino acid residue of at least 50% of the erythropoietin proteins, said link being an amine linkage.
A method of producing an erythropoietin composition comprising the steps of:
- Constructing a lower eukaryotic host cell to attach pre-selected N-linked glycans to proteins, said pre-selected N-linked glycans comprising greater than 25 mole percent of an N-linked glycan consisting essentially of a structure selected from the group consisting of
- GlcNAc2Man3GlcNAc2Gal2NANA2;
- GlcNAc2Man3GlcNAc2Gal2;
- GlcNAc2Man3GlcNAc2;
- GlcNAc2Man3;
- GlcNAc2Man5GlcNAcGalNANA;
- GlcNAc2Man5GlcNAcGal;
- GlcNAc2Man5GlcNAc;
- and GlcNAc2Man5,
- Transforming said lower eukaryotic host cell with an vector comprising a sequence encoding erythropoietin operably linked to expression control sequences recognizable by said host cell,
- Culturing said transformed cell under conditions amenable to the expression of erythropoietin, and
- Isolating the erythropoietin.
