Abstract
The present invention concerns methods and compositions usable for diagnosing Alzheimer's disease in mammals, in particular humans. It particularly describes serum markers for Alzheimer's disease and their use in diagnostic methods. It also concerns tools and/or kits usable for implementing these methods (reagents, probes, primers, antibodies, chips, cells, etc.), their preparation and their use. The invention is usable to detect the presence or progression of Alzheimer's disease in mammals, including in the early phase, as well as for predicting the efficacy of an Alzheimer's disease treatment.
Claims20
A method for detecting the presence or the risk of developing Alzheimer's disease in a mammal, or for discriminating patients with Alzheimer's from subjects with mild cognitive impairment or control subjects, comprising contacting, under conditions allowing hybridization between complementary sequences, nucleic acids from a blood sample from said mammal and a probe set, the probe set comprising at least one probe comprising all or part of each of the nucleic acid sequences of one of the following panels, or their complementary strand:
- set 1:
- 6PS,
- set 2:
- 27PS,
- set 3:
- 18PS, or
- set 4:
- 33 PS
- in order to obtain a hybridization profile, the hybridization profile being characteristic of the presence or risk of developing Alzheimer's disease in said mammal.
A method for detecting the risk of developing Alzheimer's disease in a mammal with mild cognitive impairment, or an asymptomatic one, or for discriminating patients with Alzheimer's from subjects with mild cognitive impairment, comprising contacting, under conditions allowing hybridization between complementary sequences, nucleic acids from a blood sample from said mammalian and a probe set, the probe set comprising at least one probe comprising all or part of each of the nucleic acid sequences of one of the following panels, or their complementary strand:
- set 1:
- 6PS, or
- set 2:
- 27PS
- in order to obtain a hybridization profile, the hybridization profile being characteristic of the presence or risk of developing Alzheimer's disease in said mammal.
Product comprising a support on which is immobilized a set of (groups of) probes comprising several (groups of) probes, the set containing at least one probe (or group of probes) comprising all or part of each of the nucleic acid sequences of one of the following panels, or their complementary strand:
- set 1:
- 6PS,
- set 2:
- 27PS,
- set 3:
- 18PS, or
- set 4:
- 33PS.
Product comprising a support on which at least two distinct nucleic acid probes are immobilized containing a sequence complementary and/or specific for at least two distinct target nucleic acids comprising at least two sequences chosen from among SEQ ID NO:
- 1-5578.
The use of a set of (groups of) probes, the set comprising several (groups of) probes, the set comprising at least one probe (or group of probes) comprising all or part of each of the nucleic acid sequences of any one of the following panels, or their complementary sequence:
- set 1:
- 6PS, or
- set 2:
- 27PS
- to detect in vitro or ex vivo the presence or the risk of developing Alzheimer's disease in a subject with mild cognitive impairment, or an asymptomatic one, or to discriminate AD patients from patients with mild cognitive impairment.
The use of a set of (groups of) probes comprising several (groups of) probes, the set comprising at least one probe (or group of probes) comprising all or part of each of the nucleic acid sequences of the following panel, or their complementary sequence:
- set 3:
- 18PS
- to detect in vitro or ex vivo the presence or risk of developing Alzheimer's disease in a subject or to discriminate AD patients from control patients.
