Abstract
The present invention relates to methods of distinguishing between glutamine formed by cataplerosis or proteolysis. Sample from a subject may be assayed for deuteriated glutamine (following administration of deuteriated water (2H2O) and an agent that promotes excretion of glutamine from the subject). The methods are useful in clinical settings (e.g. to test whether or not patients are suffering from proteolysis or whether athletes are abusing anabolic drugs); and may be adapted for screening test compounds for cataplerotic or proteolytic activity.
Claims17
A method of distinguishing between glutamine formed by cataplerosis or proteolysis in a subject comprising assaying a bodily sample from the subject for deuteriated glutamine
- wherein the bodily sample is from a subject that has had deuteriated water (2H2O) administered to it and has been treated with an agent that promotes excretion of glutamine from the subject;
- and
- wherein the presence of deuteriated glutamine, or a derivative thereof, indicates the proportions of glutamine in the bodily sample formed from cataplerosis and proteolysis.
A method of screening a compound to establish whether the compound is proteolytic or anabolic comprising assaying a bodily sample from a subject for deuteriated glutamine;
- wherein the bodily sample is from a subject that has had deuteriated water (2H2O) administered to it, has been treated with an agent that promotes excretion of glutamine from the subject, and been treated with the compound;
- and
- wherein, when compared to a control, an increase in the proportion of deuteriated glutamine indicates that the compound is anabolic or reduces proteolysis and a decrease in the proportion of deuteriated glutamine indicates that the compound promotes proteolysis.
