Abstract
The present invention relates to a splice variant of SIVA, SIVA3, and to its use.
Claims24
An isolated polynucleotide selected from a group consisting of (a) a polynucleotide encoding SIVA3 comprising the amino acid sequence set forth in SEQ ID NO:
- 1;
- or a derivative thereof;
- (b) a polynucleotide comprising a sequence capable of hybridizing under moderately stringent conditions to a polynucleotide sequence comprising nucleotides 119 to 324 of SEQ ID NO:
- 2 or to a fragment thereof;
- (c) a polynucleotide comprising at least 10, 11-29, 30-50, and 50-199 consecutive nucleotides, and a fragment of 200-205 consecutive nucleotides from a polynucleotide sequence comprised from 119 to 324 of SEQ ID NO:
- 2;
- and a polynucleotide of SEQ ID NO:
- 2.
(canceled)
An antibody prepared using a SIVA3 polypeptide wherein the antibody is capable of binding the SIVA3 polypeptide comprising amino acids 40 to 108 of SEQ ID NO:
- 1.
A method of producing a SIVA3 polypeptide comprising generating a transgenic animal encoding a SIVA3 polypeptide comprising amino acids 40 to 108 of SEQ ID NO:
- 1 and isolating the SIVA3 polypeptide produced by the transgenic animal from the body fluids of the transgenic animal.
12-17. (canceled)
A method for the diagnosis of a proliferative disorder or an immune disorder in a subject comprising detecting an altered level of SIVA3 in the biological sample obtained from the subject compared to the level of SIVA3 in a healthy subject, wherein said altered level is diagnostic of the proliferative disorder or the immune disorder in the subject.
A method for the follow up of a therapy for a proliferative disorder or an immune disorder in a patient comprising (a) detecting level of SIVA3 in a sample obtained from the patient before and a sample obtained from the patient after taking therapy and (b) comparing the level of SIVA3 in the patient before and after taking the therapy, wherein a change in the level of SIVA3 after taking the therapy is indicative of the efficiency of the therapy in the patient.
A method of screening for a peptide or a small molecule agonistic or antagonistic to SIVA3, comprising screening and selecting a molecule able to inhibit SIVA3 mediated NIK degradation;
- NIK mediated NF-κB activation;
- and interaction of SIVA3 polypeptide with cellular inhibitor of apoptosis protein 1 (cIAP1), or a TNF-receptor associated adaptor protein 2 (TRAF2).
A method of gene therapy for treatment of an immune disease or cancer comprising inducing the expression of a SIVA3 polypeptide or at a desired site in a human patient in need.
A clone harboring the DNA SEQ of SIVA3 deposited at the CNCM under deposit No. 1-3880.
A method for treating a disease, disorder or condition selected from developmental disorders;
- cell proliferative disorders;
- genetic disorders;
- nervous system disorders;
- metabolic disorders;
- infections and other pathological conditions;
- immune disorders;
- wound healing;
- dermatological disease;
- and ageing comprising administering to an individual affected with said disease, disorder or condition pharmaceutically acceptable composition comprising one or more active agents selected from the group consisting of a SIVA3 polypeptide or salt thereof;
- SIVA3 polynucleotide sequence;
- a vector encoding a polynucleotide encoding SIVA3;
- a host cell comprising a vector encoding a polynucleotide encoding SIVA3;
- and an antibody against SIVA3.
