Abstract
The invention provides a compound for use in the treatment of symptoms associated with psychosis, schizophrenia, a cognitive impairment disorder, a mood disorder, a nervous system disorder and/or a behavioral disorder. The compound has the formula ? and is administered to a subject in need thereof in an effective amount. The compound Meparfynol (3-methylpent-1-yn-3-ol) has been found to be effective.
Claims33
A compound of the Formula I:
- RR′R″CX
- In which:
- R is a lower alkyl of from 1-9 carbon atoms, preferably 1-5 carbon atoms or a hydroxy lower alkyl of from 1-9 carbon atoms, preferably 2-5 carbon atoms;
- R′ is a lower alkyl of from 1-9 carbon atoms, preferably 1-5 carbon atoms, or a halo such as fluoro, chloro or bromo;
- R″ is a lower alkyl of from 1-9 carbon atoms, preferably 1-5 carbon atoms, lower alkenyl of from 2-9 carbon atoms, preferably 2-5 carbon atoms, lower alkynyl of from 2-9 carbon atoms, preferably 2-5 carbon atoms and, when R and R′ are hydrogen or lower alkyl, R″ is phenyl;
- and
- X is a hydroxy, —NHCONH2, —ONHCONH2, —OCONH2 or —CONHCONH2 p1 or a pharmaceutically acceptable salt thereof or mixtures thereof or mimetics or derivatives thereof for use in the treatment of symptoms associated with psychosis, schizophrenia, a cognitive impairment disorder, a mood disorder, a nervous system disorder and/or a behavioural disorder in a subject in need of such treatment.
Use of a compound of the Formula I:
- RR′R″CX
- In which:
- R is a lower alkyl of from 1-9 carbon atoms, preferably 1-5 carbon atoms or a hydroxy lower alkyl of from 1-9 carbon atoms, preferably 2-5 carbon atoms;
- R′ is a lower alkyl of from 1-9 carbon atoms, preferably 1-5 carbon atoms, or a halo such as fluoro, chloro or bromo;
- R″ is a lower alkyl of from 1-9 carbon atoms, preferably 1-5 carbon atoms, lower alkenyl of from 2-9 carbon atoms, preferably 2-5 carbon atoms, lower alkynyl of from 2-9 carbon atoms, preferably 2-5 carbon atoms and, when R and R′ are hydrogen or lower alkyl, R″ is phenyl;
- and
- X is a hydroxy, —NHCONH2, —ONHCONH2, —OCONH2 or —CONHCONH2
- or a pharmaceutically acceptable salt thereof or mixtures thereof or mimetics or derivatives thereof in the manufacture of a medicament for use in the treatment of symptoms associated with psychosis, schizophrenia, a cognitive impairment disorder, a mood disorder, a nervous system disorder and/or a behavioural disorder in a subject in need of such treatment.
A composition in a unit dosage form comprising (a) Meparfynol;
- (b) one or more additional agents selected from the group consisting of:
- (i) an NMDA/AMPA receptor modulator;
- (ii) a nootropic agent;
- (iii) an anti-depressive agent (eg SRI);
- (iv) an Agonist/Antagonist/Partial Agonist/Partial Antagonist;
- (v) an anti-epileptic agent;
- (vi) a neuroleptic/neuropsychiatric agent;
- (vii) an atypical anti-psychotic agent;
- (viii) a cholinergic enhancer;
- and (ix) an attention stimulant/non-stimulant;
- and (c) a buffer solution wherein Meparfynol and the agent are each present at an amount of from about 10 to 7000 mg, preferably 10 mg to about 1500 mg or more, more preferably from about 50 to 600 mg.
A method of alleviating one or more symptoms associated with psychosis, schizophrenia, a cognitive impairment disorder, a mood disorder, a nervous system disorder and/or a behavioural disorder in a subject in need of such treatment wherein the method comprises by administering to an individual an effective amount of Meparfynol or a pharmaceutically acceptable salt thereof or a mixture thereof or a derivative/mimetic/analog thereof.
A method of alleviating one or more symptoms associated with sensory processing impairment in a subject in need of such treatment wherein the method comprises administering to an individual an effective amount of Meparfynol or a pharmaceutically acceptable salt thereof or a mixture thereof or a derivative/mimetic/analog thereof.
A method of alleviating one or more symptoms associated with cognitive impairment in a subject in need of such treatment wherein the method comprises administering to an individual an effective amount of Meparfynol or a pharmaceutically acceptable salt thereof or a mixture thereof or a derivative/mimetic/analog thereof.
A method of alleviating one or more symptoms associated with sensory processing impairment and cognitive impairment in a subject in need of such treatment wherein the method comprises by administering to an individual an effective amount of Meparfynol or a pharmaceutically acceptable salt thereof or a mixture thereof or a derivative/mimetic/analog thereof.
A compound of the Formula II (3-methylpent-1-yn-3-ol) as depicted in FIG. 8A or a pharmaceutically acceptable salt thereof or mixtures thereof for use in the treatment of symptoms associated with psychosis, schizophrenia, a cognitive impairment disorder, a mood disorder, a nervous system disorder and/or a behavioural disorder in a subject in need of such treatment.
A pharmaceutical composition comprising a pharmaceutically acceptable carrier and, as an active ingredient, an effective amount of 3-methylpent-1-yn-3-ol for use in the treatment of symptoms associated with psychosis, schizophrenia, a cognitive impairment disorder, a mood disorder, a nervous system disorder and/or a behavioural disorder.
