Abstract
The present invention concerns methods and reagents useful in modulating adenosine A1 receptor (ADORA1) gene expression in a variety of applications, including use in therapeutic, diagnostic, target validation, and genomic discovery applications. Specifically, the invention relates to small interfering RNA (siRNA) molecules capable of mediating RNA interference (RNAi) against ADORA1 and related receptors.

1-20. (canceled)
A chemically modified short interfering ribonucleic acid (siRNA) molecule, wherein:
- (a) the siRNA molecule comprises a sense strand and a separate antisense strand;
- (b) each strand of the siRNA is independently 18 to 24 nucleotides in length, and the siRNA comprises between 17 and 23 base pairs;
- (c) one strand of the siRNA is complementary to a human ADORA1 RNA sequence;
- (d) 10 or more pyrimidine nucleotides of the sense and antisense strand are 2′-deoxy, 2′-O-methyl, or 2′-deoxy-2′-fluoro pyrimidine nucleotides;
- (e) the siRNA molecule comprises 1, 2, 3, 4, 5, 6, 7, 8, 9, or 10 phosphorothioate internucleotide linkages;
- and,
- (f) the sense strand of the siRNA molecule comprises a terminal cap molecule at the 3′-end, the 5′-end, or both of the 3′- and 5′-ends.
