Abstract
Newly identified mammalian taste-cell-specific G Protein-Coupled Receptors and the genes encoding said receptors are described. Specifically, T2R taste G Protein-Coupled Receptors that are believed to be involved in bitter taste sensation, and the genes encoding the same, are described, along with methods for isolating such genes and for isolating and expressing such receptors. Methods for representing taste perception of a particular tastant in a mammal are also described, as are methods for generating a novel molecules or combinations of molecules that elicit a predetermined taste perception in a mammal, and methods for simulating one or more tastes.

1-137. (canceled)
n isolated or recombinant nucleic acid sequence encoding bitter taste receptor polypeptide that specifically binds and/or functionally responds to bitter ligands wherein isolated or recombinant nucleic acid sequence selected from (i) an isolated nucleic acid sequence having the nucleic acid sequence of SEQ ID NO:1;
- (ii) a nucleic acid sequence that encodes the bitter taste receptor polypeptide in SEQ ID NO:2;
- (iii) an isolated nucleic acid sequence that encodes a human bitter taste receptor that hybridizes under stringent hybridization condition to the complement of the nucleic acid sequence of SEQ ID NO:1 wherein stringent hybridization conditions are hybridization in 5×SSC, 1% SDS, incubation at 65 degrees C. and wash in 2×SSC and 0.1% SDS at 65 degrees C., and (iv) a nucleic acid sequence that encodes a bitter taste receptor polypeptide having at least 90% sequence identity to the bitter taste receptor polypeptide of SEQ ID NO:2 or to a polypeptide comprising at least all the extracellular region and/or extracellular or the transmembrane region domains of the taste receptor polypeptide in SEQ ID NO:2;
- and wherein the polypeptide encoded by the isolated or recombinant nucleic acid sequence of (i), (ii) or (iii) encodes a bitter taste receptor polypeptide that specifically binds and/or functionally responds to bitter ligands that bind or activate the bitter taste receptor polypeptide in SEQ ID NO:2.
