Abstract
Treatment of picornavirus infection by inhibiting miR-141 activity. Also disclosed herein are a method for identify miR-141 inhibitory compounds and a method for identifying a target viral infection to be treated by anti-miR-141 therapy.
Claims26
A method of treating infection caused by a picornavirus, comprising administering to a subject in need thereof an effective amount of a miR-141 inhibitory agent selected from the group consisting of an antisense oligonucleotide of miR-141 and an interfering RNA targeting early growth response 1 (EGR1).
A method of reducing viral protein production, comprising administering to a subject infected with a picornavirus an effective amount of a miR-141 inhibitory agent selected from the group consisting of an antisense oligonucleotide of miR-141 and an interfering RNA targeting early growth response 1 (EGR1).
A method for identifying an anti-picornarvirus drug candidate, comprising
- providing cells infected with a picornavirus, wherein the cells exhibit a lower level of eIF-4E relative to non-infected cells,
- contacting the cells with a compound,
- examining the activity of miR-141 in the cells, and
- determining whether the compound is a drug candidate for treating infection caused by the picornavirus,
- wherein a decrease of the miR-141 activity in the cells after the contacting step indicates that the compound is the drug candidate.
A method of identifying a target viral infection to be treated in anti-miR-141 therapy, comprising:
- providing cells infected with a virus,
- examining the level of miR-141 in the cells or the expression level of a target gene of miR-141, and
- determining whether the infection caused by the virus is a target viral infection to be treated in anti-miR-141 therapy;
- wherein a post-infection increase in the miR-141 level or a post-infection increase in the expression level of the target gene indicates that the infection is the target viral infection.
An isolated nucleic acid, comprising a nucleotide sequence at least 90% identical to a sequence selected from SEQ ID NO:1, the sequence complementary to EGR1 Region 1, the sequence complementary to EGR1 Region 2, and the sequence complementary to EGR1 Region 3.
