Pyrimidinones as Casein Kinase II (CK2) Modulators

A compound of according to Formula I:

• or a pharmaceutically acceptable salt thereof, wherein:
• X is O or S;
• R1 is —(C6-C10)aryl substituted with 1, 2 or 3 groups independently selected from —O(C1-C6)alkyl-R4, —N(H)(C1-C6)alkyl-R5, —N[(C1-C6)alkyl](C1-C6)alkyl-R6;
• —O(C1-C6)alkenyl-R7, —O(C1-C6)alkynyl-R8, —OH, —OCF3, —(C1-C6)alkyl and halo;
• R2 is benzodioxolyl, benzofuranyl, imidazolyl, 1,2-dihydro-2H-benzimidizol-2-one, 1,3-dihydro-2H-benzimidizol-2-one, thiazolyl, indazolyl, furanyl, or benzisoxazolyl, wherein each of said benzodioxolyl, benzofuranyl, imidazolyl, 1,2-dihydro-2H-benzimidizol-2-one, 1,3-dihydro-2H-benzimidizol-2-one, thiazolyl, indazolyl, furanyl, or benzisoxazolyl is optionally substituted with 1, 2 or 3 groups, which can be the same or different, selected from —N(H)(C1-C6)alkyl-N(H)C(O)—(C1-C6)alkyl-N[(C1-C6)alkyl]2, —N(H)(C1-C6)alkyl-R9, —(C1-C6)alkyl, —N[(C1-C6)alkyl](C1-C6)alkyl-R10, —N(H)C(O)(C1-C6)alkyl-N[(C1-C6)alkyl]2, —O—(C1-C6)alkyl, —NH2, —N(H)(C1-C6)alkyl-N([C1-C6)alkyl]2, —N(H)C(O)—(C1-C6)alkyl, phenyl optionally substituted with halo, —N(H)(C1-C6)alkyl-(C6-C10)aryl optionally substituted with halo or alkoxy at any ring position, and —N(H)(C1-C6)alkyl-NH2;
• R3 is H;
• or R1 and R3 can join to form a ring of 5-6 carbon atoms;
• or R1 is —(C6-C10)aryl and R2 is indazolyl substituted with 1, 2 or 3 groups, which can be the same or different, selected from —N(H)(C1-C6)alkyl-N(H)C(O)—(C1-C6)alkyl-N[(C1-C6)alkyl]2, —N(H)(C1-C6)alkyl-R9, —(C1-C6)alkyl, —N[(C1-C6)alkyl](C1-C6)alkyl-R10;
• —N(H)C(O)(C1-C6)alkyl-N[(C1-C6)alkyl]2, —O—(C1-C6)alkyl, —NH2, —N(H)C(O)—(C1-C6)alkyl, phenyl optionally substituted with halo, —N(H)(C1-C6)alkyl-(C6-C10)aryl optionally substituted with halo or alkoxy at any ring position, and —N(H)(C1-C6)alkyl-NH2;
• wherein each of R4, R5, R6, R7, R8, R9, and R10 are independently selected from H, —OH, halo, —CF3, —O(C1-C6)alkyl, —(C6-C10)aryl optionally substituted with halo or alkoxy, —O—(C1-C6)alkyl-(C6-C10)aryl,
• —(C3-C10)cycloalkyl, —CN, -(5-10 membered)heteroaryl, -(4-10 membered)heterocycloalkyl, —NH2, —N(H)(C1-C6)alkyl, and —N(H)[(C1-C6)alkyl]2.
