Abstract
Imidazole-based compounds, compositions comprising them, and methods of their use for the treatment, prevention and management of inflammatory and autoimmune diseases and disorders are disclosed. Particular compounds are of formula I:

A method of treating or managing rheumatoid arthritis, which comprises administering to a patient in need thereof a therapeutically effective amount of a compound of the formula:
- wherein:
- R1 is hydrogen, alkyl or aryl;
- R3 is OR3A, NHC(O)R3A, NHSO2R3A or hydrogen;
- each R3A is independently hydrogen or alkyl, aryl, alkylaryl, arylalkyl, heteroalkyl, heterocycle, alkylheterocycle, or heterocyclealkyl optionally substituted with halo;
- R6 is OR6A or OC(O)R6A;
- R7 is OR7A or OC(O)R7A;
- R8 is OR8A or OC(O)R8A;
- R9 is hydrogen, CH2OR9A or CH2OC(O)R9A;
- and
- each of R6A, R7A, R8A and R9A is independently hydrogen or lower alkyl.
A method of treating or managing rheumatoid arthritis, which comprises administering to a patient in need thereof a therapeutically effective amount of a compound of the formula:
- or a pharmaceutically acceptable salt thereof, wherein:
- R1 is lower alkyl;
- R3 is hydrogen, OR3A, NHC(O)R3A or NHSO2R3A;
- R9 is hydrogen or CH2OH;
- and
- each R3A is independently hydrogen or lower alkyl.
The method of claim 12, wherein the compound is of the formula:
A method of treating or managing rheumatoid arthritis, which comprises administering to a patient in need thereof a therapeutically effective amount of (E)-1-(4-((1R,2S,3R)-1,2,3,4-tetrahydroxybutyl)-1H-imidazol-2-yl)-ethanone oxime.
