Dutpase inhibitors

A compound of the formula I

• where
• A is O, S or CH2;
• B is O, S or CHR3;
• R1 is H, C1-C5 alkyl, C2-C5 alkenyl, C2-C5 alkynyl or a 5 or 6 membered, saturated or unsaturated ring containing 0 to 3 heteroatoms selected from N, O and S, the alkyl, alkenyl, alkynyl or ring being independently optionally substituted with R4;
• R2 is H, F;
• R3 is H, F, OH, NH2 or a pharmaceutically acceptable ester, amide or ether thereof;
• or
• R2 and R3 together form a chemical bond;
• D is ONHCO—, —CONH—, —O—, —C(═O)—, —CH═CH, —C═C—, —NR5—;
• R4 is independently selected from hydrogen, halo, cyano, amino, nitro, carboxy, carbamoyl, hydroxy, oxo, C1-C5 alkyl, C1-C5 haloalkyl, C1-C5 alkyloxy, C1-C5 alkanoyl, C1-C5 alkanoyloxy, C1-C5 alkylthio, —N(C0-C3-alkyl)2, hydroxymethyl, aminomethyl, carboxymethyl;
• —SOnN(C0-C3-alkyl), —SOnC1-C5-alkyl, where n is 1 or 2;
• R5 is H, C1-C4-alkyl, C1-C4-alkanoyl;
• E is Si or C;
• R6 and R7 are independently a stable monocyclic, bicyclic or tricyclic ring system which has an aromatic nature and wherein each ring has 0 to 3 heteroatoms selected from N, O and S;
• R8 is C1-C8 alkyl, C2-C8 alkenyl, C2-C8 alkynyl, or a stable monocyclic, bicyclic or tricyclic ring system which is saturated or unsaturated and in which each ring has 0 to 3 heteroatoms selected from N, O and S;
• R6, R7 and R8 are independently optionally substituted with R4;
• with the proviso that if the group C0-C3alkyl-D-C0-C3 alkyl is —O—CH2—, then the group E(R6)(R7)(R8) is not CPh3 (trityl), methoxylated trityl or tert.butyldiphenylsilyl;
• and pharmaceutically acceptable salts thereof.
