Abstract
The invention provides methods of coupling protein ligands to a solid support. The invention also provides affinity chromatography matrices and methods of using affinity chromatography matrices to purify a target molecule.
Claims25
An affinity chromatography matrix comprising:
- a) a solid support;
- b) a protein ligand bound to the solid support;
- and c) an associative group separately bound to the solid support, wherein the associative group interacts non-covalently with the protein ligand.
A method of purifying a target molecule from a mixture comprising:
- 1) contacting the mixture under a first set of conditions with an affinity chromatography matrix such that the target molecule binds to the protein ligand of the matrix, wherein the affinity chromatography matrix comprises:
- a) a solid support;
- b) a protein ligand bound to the solid support;
- and c) an associative group separately bound to the solid support where the associative group interacts with the protein ligand;
- and 2) altering the conditions of 1) such that the target molecule no longer binds to the protein ligand of the matrix, thereby purifying the substance of interest.
2 more elements
An affinity chromatography matrix comprising:
- a) an agarose support;
- b) protein A covalently bound to the agarose support;
- and c) a charged species, which separately and covalently binds with the solid support and non-covalently interacts with the protein A.
An agarose bead comprising at least 10 g/L of protein A bound to the surface of the agarose bead.
