Abstract
A recombinant isolated antibody and a pharmaceutical composition containing same, capable of specifically recognizing an MHC-peptide complex with an affinity in a nanomolar range and of inducing apoptosis in cancer or pathogen infected cells is provided. Also provided are method for treating and diagnosing cancer or pathogen infection in a subject using the recombinant isolated antibody of the present invention.
Claims30
A method of inducing apoptosis in cancer cells or pathogen infected cells, the method comprising contacting, or expressing in the cells a recombinant isolated antibody capable of specifically binding an MHC-peptide complex specifically expressed on the cells, thereby inducing apoptosis in the cancer cells or the pathogen infected cells.
A method of treating cancer or pathogen infection comprising administering to, or expressing in cells of a subject in need thereof a therapeutically effective amount of a recombinant isolated antibody capable of specifically binding an MHC-peptide complex, said recombinant isolated antibody being capable of inducing apoptosis of cancer cells or pathogen infected cells, said cancer cells or said pathogen infected cells specifically expressing said MHC-peptide complex, thereby treating the cancer or the pathogen infection in the subject.
Use of a recombinant isolated antibody capable of specifically binding an MHC-peptide complex, as a pharmaceutical, said recombinant isolated antibody being capable of inducing apoptosis of cancer cells or pathogen infected cells specifically expressing said MHC-peptide complex.
Use of a recombinant isolated antibody capable of specifically binding an MHC-peptide complex, said recombinant isolated antibody being capable of inducing apoptosis of cancer cells or pathogen infected cells specifically expressing said MHC-peptide complex, for the manufacture of a medicament identified for the treatment of cancer or pathogen infection.
A pharmaceutical composition for treating cancer or pathogen infection comprising as an active ingredient a recombinant isolated antibody capable of specifically binding an MHC-peptide complex in cancer cells or pathogen infected cells specifically expressing said MHC-peptide complex, said recombinant isolated antibody being capable of inducing apoptosis in said cancer cells or said pathogen infected cells and a pharmaceutically acceptable carrier.
A method of diagnosing cancer or pathogen infection in a subject, comprising:
- (a) contacting a biological sample of the subject with a recombinant isolated antibody capable of specifically binding an MHC-peptide complex under conditions suitable for immunocomplex formation;
- said recombinant isolated antibody being capable of inducing apoptosis in target cancer cells or pathogen infected cells, and
- (b) detecting formation of said immunocomplex, thereby diagnosing the cancer or pathogen infection in the subject.
A method of diagnosing cancer or pathogen infection in a subject, comprising:
- (a) contacting a biological sample of the subject with a recombinant isolated antibody capable of specifically binding an MHC-peptide complex under conditions suitable for immunocomplex formation, said recombinant isolated antibody being capable of inducing apoptosis in target cancer cells or pathogen infected cells;
- and
- (b) detecting a level of apoptosis in cells of said biological sample thereby diagnosing the cancer or pathogen infection in the subject.
A kit for diagnosing cancer or pathogen infection in a subject, the kit comprising packaging materials and at least one agent identified by said packaging materials as being suitable for an immunocomplex formation between a recombinant isolated antibody capable of specifically binding an MHC-peptide complex and said MHC-peptide complex, said recombinant isolated antibody being capable of inducing apoptosis in cancer cells or pathogen infected cells.
A recombinant antibody capable of inducing apoptosis in cancer cells or pathogen infected cells.
A pharmaceutical composition comprising a recombinant antibody capable of inducing apoptosis in cancer cells or pathogen infected cells, and a pharmaceutical acceptable carrier.
A recombinant 9H scFv as set forth by SEQ ID NO:1.
A recombinant CLA12 scFv as set forth by SEQ ID NO:17.
A recombinant antibody comprising at least one CDR selected from the group consisting of SEQ ID NOs:5-16 and 19-24.
