Abstract
Apparatus and methods for amplification of DNA are provided that use sonic energy in place of conventional thermocyclers. In one embodiment, sonic energy is applied to a PCR cocktail to effect dissociation of double stranded DNA into single strands of DNA. A quiescence stage, where no sonic energy is applied, results in amplification of DNA. Repetition of the cycles of application of sonic energy and cessation of application of sonic energy results in DNA amplification.
Claims25
A cycler for DNA amplification, comprising:
- a sonic energy source suitable for dissociating double stranded DNA into single stranded DNA without raising the temperature of the double stranded DNA to a temperature at which thermal denaturation will occur;
- a controller suitable for operating the sonic energy source when it is desired to effect dissociation of double stranded DNA, and for discontinuing the operation of the sonic energy source when it is desired to allow amplification of single stranded DNA in the presence of suitable polymerase enzyme, nucleosides and primers.
A cycler for DNA amplification, comprising:
- a vial holder configured to receive at least one vial containing a reaction mixture including DNA, a polymerase enzyme, nucleosides and primers;
- a sonic energy source suitable for dissociating double stranded DNA into single stranded DNA without raising the temperature of the double stranded DNA to a temperature at which thermal denaturation will occur;
- a controller suitable for:
- operating the sonic energy source when it is desired to effect dissociation of double stranded DNA contained in a vial placed in the vial holder into single stranded DNA,
- discontinuing the operation of the sonic energy source when it is desired to allow amplification of single stranded DNA in the presence of suitable polymerase enzyme, nucleosides and primers contained in a reaction mixture;
- and
- repeating the operation of the sonic energy source and discontinuing the operation of the sonic energy source in order to produce a desired amount of DNA.
