Cysteine Protease Inhibitors

A compound of the formula II

• wherein
• one of R1 and R2 is halo and the other is H or halo;
• R3 is —C1-C5 straight or branched chain, optionally fluorinated, alkyl or —CH2CR5C3-C4-cycloalkyl;
• R4 is H;
• R5 is H, C1-C2 alkyl, C1-C2 haloalkyl, hydroxyl, OC1-C2alkyl, fluoro;
• R6 is a stable, optionally substituted, monocyclic or bicyclic, carbocycle or hetorocycle wherein the or each ring has 4, 5 or 6 ring atoms and 0 to 3 hetero atoms selected from S, O and N and wherein the optional substituents comprise 1 to 3 members selected from R7;
• R7 is independently selected from halo, oxo, nitrite, nitro, C1-C4 alkyl, —XNRdRe, —XNReR8, —NReXR8, NH2CO—, X—R8, X—O—R8, O—X—R8, X—C(═O)R8, X(C═O)NRdR8, X—NReC(═O)R8, X—NHSOmR8, X—S(═O)n, R8, X—C(═O)OR8, XNReC(═O)OR8;
• R8 is independently H, C1-C4 alkyl, C3-C6 cycloalkyl, pyrrolidinyl, piperidinyl, morpholinyl, thiomorpholinyl, piperazinyl, indolinyl, pyranyl, thiopyranyl, furanyl, thienyl, pyrrolyl, oxazolyl, isoxazolyl, thiazolyl, imidazolyl, pyridinyl, pyrimidinyl, pyrazinyl, indolyl, phenyl, any of which is optionally substituted with up to 3 members selected from R9;
• R9 is independently selected from hydroxy, XR10, —XRdRe, —XNReR10, —NReC1-C4alkylR10, —S(═O)mRe, cyano, carboxy, oxo, C1-C4 alkyl, C1-C4haloalkyl, C1-C4-alkoxy, C1-C4 alkanoyl, carbamoyl;
• R10 is C3-C6 cycloalkyl, pyrrolidinyl, piperidinyl, morpholinyl, thiomorpholinyl, piperazinyl, indolinyl, pyranyl, thiopyranyl, furanyl, thienyl, pyrrolyl, oxazolyl, isoxazolyl, thiazolyl, imidazolyl, pyridinyl, pyrimidinyl, pyrazinyl, indolyl, phenyl, any of which is optionally substituted with C1-C4 alkyl, halo, hydroxy, C1-C4alkoxy, cyano, —S(═O)mRe, C1-C4-haloalkyl;
• X is independently a bond or C1-C4 alkylene;
• Ra is independently H, C1-C4 alkyl or CH3C(═O);
• Rb is C1-C4 haloalkyl;
• Rc is H, C1-C4 alkyl;
• or Re together with R6 and the carbon atom to which they are both attached form a carbocycle or heterocycle as defined for R6;
• Rd is independently H, C1-C4 alkyl or CH3C(═O);
• Re is independently H, C1-C4 alkyl;
• or
• Rd and Re together with the N atom to which they are attached form a morpholine, piperidine, piperazine or pyrrolidine ring optionally substituted with R9;
• m is independently 0, 1 or 2;
• or a pharmaceutically acceptable salt, hydrate or N-oxide thereof.
