Abstract
Novel formulations comprising therapeutic agents, a surface tension-controlling agent, and a component that comprises both a humectant and a viscosity-controlling agent for use in thermal droplet ejecting devices are disclosed. The formulations are useful for maintaining controlled and reliable dosage of the therapeutic agent via pulmonary delivery.

A device for pulmonary delivery of a therapeutic protein to a patient, the device comprising a computer-controlled electronic aerosol generating system fluidly connected to a reservoir containing a liquid pharmaceutical composition comprising a therapeutic protein, a surface-tension controlling agent and a humectant.
A method of making a device for pulmonary delivery of a therapeutic protein to a patient, the method comprising fluidly connecting a computer-controlled electronic to aerosol generating system to a reservoir containing a liquid pharmaceutical composition comprising a therapeutic protein, a surface tension-controlling agent and a humectant.
A liquid pharmaceutical composition, said composition contained in a fluid is reservoir equipped with an electronic aerosol generating system, the composition comprising:
- a therapeutic agent,
- a surface tension-controlling agent, and
- a component comprising a humectant and a viscosity-controlling agent.
A liquid pharmaceutical composition contained in a fluid reservoir of a thermal droplet ejecting device equipped with an electronic aerosol generating system for aerosolization by the device for administration via inhalation, said composition comprising a therapeutic agent, a surface tension-controlling agent, and a component comprising a humectant and a viscosity-controlling agent, wherein the surface tension-controlling agent provides for a solution having a surface tension of 8 dynes/cm to 75 dynes/cm, and wherein the liquid pharmaceutical composition contains water and is in the form of an aqueous solution, emulsion or suspension.
A liquid pharmaceutical composition contained in a fluid reservoir of a thermal droplet ejecting device equipped with an electronic aerosol generating system for aerosolization by the device for administration by inhalation, the liquid pharmaceutical composition comprising:
- a therapeutic agent,
- a surface tension-controlling agent comprising 0.01% to 3% w/v of the composition, and
- a component comprising a humectant and a viscosity-controlling agent,
- wherein the liquid composition has a surface tension of 8 dynes/cm to 75 dynes/cm, a viscosity of 2 cp to 10 cp, and a density of 0.7 g/mL to 2.2 g/mL, and
- wherein the liquid pharmaceutical composition contains water and is in the form of an aqueous solution, emulsion or suspension.
A liquid pharmaceutical composition contained in a fluid reservoir of a thermal droplet ejecting device for aerosolization by the device for administration via inhalation, said composition comprising a therapeutic agent, a surface tension-controlling agent, a component comprising a humectant and a viscosity-controlling agent, and wherein the surface tension-controlling agent provides for a solution having a surface tension of 8 dynes/cm to 75 dynes/cm.
A liquid pharmaceutical composition contained in a fluid reservoir of a thermal droplet ejecting device, the liquid pharmaceutical composition comprising
- a therapeutic agent,
- a surface tension-controlling agent comprising 0.01% to 3% w/v of the composition, and
- a component comprising a humectant and a viscosity-controlling agent,
- wherein the liquid composition has a surface tension of 8 dynes/cm to 75 dynes/cm, a viscosity of 2 cp to 10 cp, and a density of 0.7 g/mL to 22 g/mL.
