Abstract
Disclosed is a method for the mass production of a monomeric or dimeric immunoglobulin Fc region, free of initial methionine residues, using a recombinant expression vector comprising a nucleotide sequence coding for a recombinant immunoglobulin Fc region comprising an immunoglobulin Fc region linked at the N-terminus thereof to an immunoglobulin Fc region via a peptide bond.
Claims14
A method of producing an immunoglobulin Fc region free of an initial methionine residue on a mass scale, comprising:
- preparing a recombinant expression vector including a nucleotide sequence coding for a recombinant immunoglobulin Fc region comprising an immunoglobulin Fc region linked at an N-terminus thereof to an immunoglobulin hinge region via a peptide bond;
- transforming a prokaryotic cell with the recombinant expression vector to create a transformant;
- culturing the transformant to express the immunoglobulin Fc region as an inclusion body;
- and
- isolating and purifying the immunoglobulin Fc region.
