Abstract
A method is provided for transforming maize plants to express DNA sequences of interest from plant cell plastids. The method allows the transformation of maize plant tissue with heterologous DNA constructs by microprojectile bombardment of green light grown cultures and dark grown embryogenic cultures. The invention also provides for maize cells in which the plastids contain heterologous DNA constructs.
Claims14
A method for transforming plastids in a maize cell, wherein the method comprises the steps of:
- obtaining a green multiple bud culture;
- introducing selected exogenous DNA into said green multiple bud culture, said selected exogenous DNA being on a vector capable of introducing said DNA into a plastid;
- and selecting a cell containing said selected exogenous DNA in plastids of said cell, wherein the DNA is stably integrated and inherited through organelle replication in daughter cells.
A method for transforming plastids in a maize cell comprising:
- obtaining a dark grown embryogenic callus;
- culturing said dark grown embryogenic callus under light conditions for a period of 3-10 days to obtain somatic embryos exhibiting a greening phenotype as compared to embryos grown under dark conditions;
- selecting said greening somatic embryos;
- introducing selected exogenous DNA into said greening somatic embryos, said selected exogenous DNA being on a vector capable of introducing said DNA into a plastid;
- and selecting cells containing said selected exogenous DNA in plastids of said cell, wherein the DNA is stably integrated and inherited through organelle replication in daughter cells.
