## Abstract

Acute myeloid leukemia stem cells (AMLSC) are identified. The cells can be prospectively isolated or identified from patient samples, and are shown to possess the unique properties of cancer stem cells in functional assays for cancer stem cell self-renewal and differentiation, and in cancer diagnosis.

## Claims

### Claim 1

A method for characterizing an acute myeloid leukemia from a patient, the method comprising:
contacting a patient sample with reagents specific for CD96;
quantitating the number of CD96+ cancer cells,
wherein the presence of CD96+ cancer cells is indicative of the presence of AML cancer stem cells.

### Claim 2 (depends on 1)

The method according to claim 1, wherein said reagents include reagents specific for CD96.

### Claim 3 (depends on 1)

The method of claim 1, wherein the patient sample is contacted with reagents specific for CD34 and for CD38, wherein AML cancer stem cells are CD34+CD38−.

### Claim 4 (depends on 1)

The method of claim 1, wherein the quantitating is performed by flow cytometry.

### Claim 5 (depends on 1)

The method of claim 1, wherein the quantitating is performed by immunohistochemistry.

### Claim 6 (depends on 1)

The method of claim 1, wherein the sample is a blood sample.

### Claim 7 (depends on 1)

The method of claim 1, wherein the patient has been diagnosed as having AML.

### Claim 8 (depends on 7)

The method of claim 7, wherein the patient is undergoing treatment for AML.

### Claim 9 (depends on 1)

The method according to claim 1, wherein said patient is a human.

### Claim 10

A composition of mammalian acute myeloid leukemia stem cells, wherein at least 50% of the cells in said composition are CD96+, CD34+, CD38− acute myeloid leukemia stem cells (AMLSC).

### Claim 11 (depends on 10)

The composition according to claim 10, wherein at least 75% of the cells in said composition are AMLSC.

### Claim 12 (depends on 11)

The composition according to claim 11, wherein said AMLSC are human cells.

### Claim 13 (depends on 1, 2, 3, 4, 5, 6, 7, 8, 9)

A kit for use in any of the methods set forth in claims 1-9.

### Claim 14 (depends on 10)

A method of screening a candidate chemotherapeutic agent for effectiveness against an AMLSC, the method comprising:
contacting said agent with the cell composition of claim 10, and
determining the effectiveness of said agent against said AMLSC.

### Claim 15

A method of targeting or depleting AML cancer stem cells, the method comprising contacting reagent blood cells with an agent that specifically binds CD96 in order to target or deplete CD96+ AMLSC.
