Abstract
This invention relates to transgenically produced human Antithrombin III (tgATIII). The human ATIII produced by the transgenic process of the present invention has a monosaccharide composition which comprises N-acetylgalactosamine (GalNAc) along with fucose, N-acetylglucosamine, galactose, mannose, and N-acetylneuraminic acid/N-glycolyneuraminic acid. The monosaccharide composition differs with that of plasma derived ATIII (phATIII). It has been found that tgATIII has an increased clearance rate when compared to phATIII.
Claims7
1-11. (canceled)
A method of treating a subject having an antithrombin III (ATE) deficiency or inflammation, comprising:
- administering to a patient a non-human mammal mammary gland produced recombinant antithrombin III (ATIII).
The method of claim 1, wherein the mammary gland produced antithrombin III (ATE) is produced in the mammary glands of a transgenic goat.
The method of claim 1, wherein said non-human mammal is produced by microinjection procedures.
The method of claim 1, wherein said antithrombin purification is at least 90% pure as determined by ELISA.
The antithrombin III (ATIII) as produced by the method of claim 1 further comprising antithrombin III (ATIII) whose glycosylation varies from plasma-derived antithrombin III (ATIII) through the increased presence of GalNAc and reduction of Sialic Acid.
The antithrombin III (ATIII) of claim 1 produced at least 5 grams liter.
