Abstract
The present invention relates to reagents and methods for the detection of osteopontin fragments and distinguishing them from each other and from the full-length osteopontin protein. The present invention also relates to assays for the determination of the presence of osteopontin fragments in samples obtained from subjects and, further, the correlation of osteopontin fragment levels fragment levels with disease detection, progression and prognosis.
Claims19
A method of identifying a fragment of osteopontin, the method comprising:
- a) capturing the osteopontin fragment with a capture antibody which binds specifically to a first epitope of the osteopontin fragment;
- b) detecting the captured osteopontin fragment with an antibody which binds specifically to a second epitope of the osteopontin fragment;
- and
- c) determining that the peptide is a fragment by detecting a lack of binding to osteopontin by a determining antibody to a third epitope, as compared, to a control sample comprising the full length osteopontin peptide.
A method of identifying a fragment of osteopontin, the method comprising:
- a) capturing the osteopontin fragment with a capture antibody which binds specifically to a either the N-terminus or C-terminus of the osteopontin protein;
- and,
- b) determining that the peptide is a fragment by determining a lack of binding to osteopontin by a determining antibody which binds specifically either the C-terminus of the osteopontin protein, if the capture antibody was binds specifically to the N-terminus, or by a determining antibody which binds specifically to the N-terminus, if the capture antibody binds specifically to the C-terminus, as compared to a control sample comprising the full length osteopontin peptide.
