Abstract
An effective cancer cell vaccine for canines has been developed. The vaccine is prepared from autologous lymphoma cells transfected with emm55. Once an animal is vaccinated, the expressed Emm55 antigen stimulates an immunogenic response to the tumor cells resulting in significantly increased survival, strong autologous and cross reactive humoral and cell mediated responses in several breeds of dogs diagnosed with later stage lymphomas.
Claims18
A method for treating a lymphoma, comprising:
- obtaining a lymphoma tissue sample;
- transforming lymphoma cells from the tissue sample with a gene encoding Emm55;
- administering the transformed lymphoma cells to the subject in an amount effective to adversely affect tumor proliferation in the subject.
A composition comprising Emm55 gene transformed autologous or allogeneic lymphoma cells.
An autologous or allogeneic lymphoma cell transformed with an expression vector comprising a nucleic acid encoding a Streptococcus pyogenes Emm55 polypeptide.
An autologous or allogeneic lymphoma cell population modified to express a Streptococcus pyogenes Emm55 polypeptide comprised within a pharmaceutically acceptable vehicle suitable administration to a subject exhibiting a lymphoma type tumor.
A lymphoma cell vaccine comprising a population of autologous or allogenic mammalian lymphoma cells transformed with an expression vector containing a nucleic acid encoding Streptococcus pyogenes Emm55 polypeptide.
A method of preparing a lymphoma cancer cell vaccine, comprising:
- isolating autologous or allogeneic lymphoma cells;
- transfecting the cells with a plasmid vector containing Streptococcus pyogenes emm55 gene and a drug resistance gene;
- selecting cells resistant to said drug;
- expanding the drug selected cells;
- irradiating the expanded cells;
- and
- preparing a lymphoma cancer cell vaccine from the irradiated cells.
A method of in vivo transformation of one or more autologous lymphoma cells, comprising providing to said one or more cells a nucleic acid that encodes Emm55.
