Abstract
The present invention relates to a method for stabilization of analytes in solutions of solubilized body samples. The method comprises the steps of solubilizing the body samples obtained from a subject in a suitable sample medium and stabilizing said body sample contained within the sample medium by heating said sample medium for a certain period of time. A further aspect of the invention is a method of a denaturing immunoassay of proteins in solution. The method comprises the steps of bringing a sample containing proteins into contact with a denaturing agent and heating said sample in the presence of the denaturing agent to allow the protein to be denatured.
Claims23
A method for stabilization of one or more analytes in solutions of solubilized body samples comprising the steps of:
- i) solubilizing a body sample containing one or more analytes obtained from a mammalian subject in a sample medium comprising at least one denaturing agent selected from the group consisting of detergents and chaotropic agents;
- ii) subsequently heating said sample medium containing the sample to a temperature of at least 70° C. for a period of time of at least 3 minutes;
- and
- iii) stabilizing the one or more analytes that have been solubilized within the sample medium for storage and transport and subsequent immunoassay.
A method for detecting one or more analyte proteins by an immunoassay, comprising the step of carrying out an immunoassay in the presence of one or more denaturing agents in a concentration to allow to uphold the denatured status of the one or more analyte proteins.
An automated heating device designed to perform the steps of:
- i) Heating at least one container comprising a solution of a solubilized body sample to a preset temperature;
- ii) Maintaining said temperature within the sample liquid for a predefined amount of time;
- and
- iii) giving an acoustic, vibration or optic signal indicating the end of the incubation period.
The heating device according to claim 22, wherein the preset temperature is in the range of 90° C. to 110° C.
The heating device according to claim 22, wherein the preset temperature is in the range of 95° C. to 99° C.
The heating device according to claim 22, wherein the preset time is in the range of 7 minutes to 30 minutes.
