Abstract
The invention provides nucleic acids, and variants and fragments thereof, obtained from strains of Bacillus thuringiensis encoding δ-endotoxins having pesticidal activity against insect pests, including Lepidoptera. Particular embodiments of the invention provide isolated nucleic acids encoding pesticidal proteins, pesticidal compositions, expression cassettes, and transformed microorganisms and plants comprising a nucleic acid of the invention. These compositions find use in methods for controlling pests, especially plant pests.
Claims7
An isolated nucleic acid comprising a nucleotide sequence that encodes a polypeptide having pesticidal activity, wherein said nucleotide sequence is selected from the group consisting of:
- (a) the nucleotide sequence set forth in SEQ ID NO:3, 26, or 30;
- (b) a nucleotide sequence encoding the amino acid sequence set forth in SEQ ID NO:4, 27, or 31;
- (c) a nucleotide sequence encoding an amino acid sequence corresponding to domain 1 of the protein encoded by SEQ ID NO:
- 4, 27, or 31;
- (d) a nucleotide sequence encoding an amino acid sequence corresponding to domain 2 of the protein encoded by SEQ ID NO:
- 4, 27, or 31;
- (e) a nucleotide sequence having at least 90% sequence identity to a nucleotide sequence of (a);
- (f) a nucleotide sequence having at least 95% sequence identity to a nucleotide sequence of (a);
- (g) a nucleotide sequence encoding a protein comprising an amino acid sequence characterized by at least 90% sequence identity to an amino acid sequence of (b);
- (h) a nucleotide sequence encoding a protein comprising an amino acid sequence characterized by at least 95% sequence identity to an amino acid sequence of (b);
- and
- (i) a nucleotide sequence comprising at least 500 contiguous nucleotides of the nucleotide sequence set forth in SEQ ID NO:
- 3, 26, or 30.
A transformed plant comprising in its genome at least one stably incorporated nucleotide construct comprising a nucleotide sequence that encodes a polypeptide that is pesticidal for at least one pest belonging to the order Lepidoptera, wherein said coding sequence is operably linked to a promoter that drives expression in a plant cell and wherein said coding sequence is selected from the group consisting of:
- (a) the nucleotide sequence set forth in SEQ ID NO:3, 26, or 30;
- (b) a nucleotide sequence encoding the amino acid sequence set forth in SEQ ID NO:4, 27, or 31;
- (c) a nucleotide sequence encoding an amino acid sequence corresponding to domain 1 of the protein encoded by SEQ ID NO:
- 4, 27, or 31;
- (d) a nucleotide sequence encoding an amino acid sequence corresponding to domain 2 of the protein encoded by SEQ ID NO:
- 4, 27, or 31;
- (e) a nucleotide sequence characterized by at least 90% sequence identity to a nucleotide sequence of (a);
- (f) a nucleotide sequence characterized by at least 95% sequence identity to a nucleotide sequence of (a);
- (g) a nucleotide sequence encoding a protein comprising an amino acid sequence characterized by at least 90% sequence identity to an amino acid sequence of (b);
- (h) a nucleotide sequence encoding a protein comprising an amino acid sequence characterized by at least 95% sequence identity to an amino acid sequence of (b);
- (i) a nucleotide sequence according to any one of (a) to (d) that comprises codons optimized for expression in a plant;
- and
- (j) a nucleotide sequence comprising at least 500 contiguous nucleotides of the nucleotide sequence set forth in SEQ ID NO:3, 27, or 31.
