Abstract
Novel Enterococcus and Streptococcus bacteriocins produced by novel Enterococcus and Streptococcus strains are used for at least reducing the levels of colonization by at least one target bacteria in animals, especially poultry.
Claims15
An isolated bacteriocin produced by an Enterococcus strain or a Streptococcus strain having the identifying characteristics of a strain selected from the group consisting of NRRL B-30746 and NRRL B-30745.
An isolated Enterococcus faecium strain having the identifying characteristics of NRRL B-30746.
An isolated Streptococcus cricetus strain having the identifying characteristics of NRRL B-30745.
A therapeutic composition comprising:
- (a) at least one isolated bacteriocin produced by an Enterococcus strain or a Streptococcus strain having the identifying characteristics of a strain selected from the group consisting of NRRL B-30746, NRRL B-3745, and mixtures thereof in an amount effective to at least reduce levels of colonization by at least one target bacteria;
- and
- (b) a suitable therapeutic carrier.
A therapeutic feed for animals comprising:
- (a) at least one isolated bacteriocin produced by an Enterococcus strain or a Streptococcus strain having the identifying characteristics of a strain selected from the group consisting of NRRL B-30746, NRRL B-30745, and mixtures thereof in amounts effective to at least reduce the levels of colonization by at least one target bacteria,
- (b) a therapeutic carrier, and
- (c) an animal feed.
The therapeutic feed of claim 11 wherein said at least one bacteriocin has an amino acid sequence selected from the group consisting of SEQ ID NO 1, SEQ ID NO 2, and mixtures thereof.
A method for at least reducing the level of colonization by at least one a target bacteria in an animal comprising:
- administering to an animal a therapeutic composition comprising at least one isolated bacteriocin produced by an Enterococcus strain or a Streptococcus strain having the identifying characteristics of a strain selected from the group consisting of NRRL B-30746, NRRL 30745, and mixtures thereof in amounts effective to at least reduce the levels of colonization by at least one target bacteria and a therapeutic carrier.
An isolated strain of Lactococcus lactis having the identifying characteristics of NRRL B-30744.
A method for increasing the production of bacteriocins by bacteria comprising:
- (a) adding a bacteriocin-enhancing amount of Lactococcus lactis having the identifying characteristics of NRRL B-30744 to a culture of a bacteriocin-producing bacteria to form a co-culture, and
- (b) culturing said co-culture at about 37° C.
A method for isolating a bacteriocin comprising:
- (a) co-culturing a bacteriocin-producing bacteria with an inducer bacteria to produce bacteriocin in a culture medium,
- (b) harvesting said culture medium and cells by centrifugation to obtain a supernatant and a cell pellet,
- (c) applying said bacteriocin-containing supernatant to hydrophobic interaction chromatography column to obtain an isolated bactericin preparation,
- (d) incubating said cell pellet from step b in an eluting buffer to form a first mixture,
- (e) separating cells from said cell pellet and said buffer by centrifugation,
- (f) isolating bacteriocin from said buffer by ion-exchange chromatography.
