Abstract
The present invention stems from the realization that the cell surface content of sialic acids is associated with drug susceptibility and resistance in neoplastic and damaged cells. A general decrease in the amounts of the α2-6 linked sialic acid has been confirmed in resistant phenotype compare to its corresponding sensitive parental cells. Treating the resistant phenotype with neuraminidase, an enzyme that removes the sialic acid from the sugar chain, facilitates the drug internalization and reinstalls the cell susceptibility to the drug. Based on these observations, methods for predicting the resistance to a number of drugs and detecting multi-drug resistanc in neoplastic and damaged cells have been invented.
Claims5
A method for determining multi-drug resistance of a cancer patient, comprising:
- determining level of sialic acids at a surface of cells of said cancer patient;
- treating, after said determining, said patient with at least one drug for said cancer;
- determining level of sialic acid at a surface of cells of said cancer patient after said treating;
- comparing levels of sialic acid before and after said treating to determine whether cells of said patient after said treating are exhibiting multi-drug resistance.
A method for determining multi-drug resistant cells of a cancer patient, comprising:
- determining level of sialic acid at a surface of cells of said patient;
- treating, after said determining, said cells of said patient with at least one drug or a series of drugs for said cancer;
- determining level of sialic acid at a surface of said cells of said cancer patient after said treating;
- comparing levels of sialic acid before and after said treating to determine whether cells of said patient after said treating are exhibiting multi-drug resistance and compare the results of different drugs to obtain the best strategy for treatment.
A method for determining multi-drug resistant cells of a cancer patient, comprising:
- determining level of sialic acid at a surface of cells of said patient;
- comparing said level of sialic acid to a statistically determined silaic level in cells to determine whether cells of said patient are exhibiting multi-drug resistance, wherein when said level of sialic acid is lower than said statistically determined silaic level said cells of said cancer patient are exhibiting a tendency to multi-drug resistance.
