Abstract
Agents for treating pain, methods for producing the agents and methods for treating pain by administration to a patient of a therapeutically effective amount of the agent. The agent can include a clostridial neurotoxin, or a component or fragment or derivative thereof, attached to a targeting moiety, wherein the targeting moiety is selected from a group consisting of transmission compounds which can be released from neurons upon the transmission of pain signals by the neurons, and compounds substantially similar to the transmission compounds.
Claims9
A polynucleotide molecule encoding a modified Clostridial toxin comprising:
- a) a first polynucleotide region comprising;
- i) a first polynucleotide segment encoding an orphanin targeting moiety capable of binding to an orphanin receptor;
- and
- ii) a second polynucleotide segment encoding a Clostridial toxin translocation domain or fragment thereof capable of translocating a Clostridial toxin light chain or light chain fragment thereof across an endosome membrane;
- and
- b) a second polynucleotide region encoding a Clostridial toxin light chain or light chain fragment thereof capable of cleaving a neurosecretory protein.
- wherein the Clostridial toxin binding domain of the modified Clostridial toxin is removed or modified in order to reduce the targeting of the modified Clostridial toxin to the neuromuscular junction.
A method for obtaining a modified Clostridial toxin, the method comprising:
- a) providing a genetic construct encoding a modified Clostridial toxin comprising a Clostridial toxin proteolytic domain;
- a Clostridial toxin translocation domain and an orphanin targeting moiety;
- wherein the Clostridial toxin binding domain of the modified Clostridial toxin is removed or modified in order to reduce the targeting of the modified Clostridial toxin to the neuromuscular junction;
- and
- b) expressing the construct in a host organism to produce the modified Clostridial toxin.
A method for obtaining a modified Clostridial toxin, the method comprising:
- a) providing a genetic construct encoding a modified Clostridial toxin comprising a Clostridial toxin proteolytic domain and a Clostridial toxin translocation;
- wherein the Clostridial toxin binding domain of the modified Clostridial toxin is removed or modified in order to reduce the targeting of the modified Clostridial toxin to the neuromuscular junction;
- b) expressing the construct in a host organism to produce the modified Clostridial toxin;
- and
- c) covalently attaching the modified Clostridial toxin to an orphanin targeting moiety.
