Abstract
The present invention provides an assay for the identification of agents which can modulate TOR-mediated phosphorylation of substrate proteins. The assays of the invention utilize substrate proteins whose amino acid sequence contains the Ser/Thr motif recognized by TOR. Naturally occurring TOR which may be used in the methods of the invention include TOR isolated from a variety of species, particularly mammalian tissues.
Claims17
A method for measuring the ability of an agent to modulate the ability of native Target of Rapamycin (TOR) protein to phosphorylate a substrate, comprising determining the level of phosphorylation of said substrate under conditions in which TOR and the substrate interact:
- (a) in the absence of said agent;
- (b) in the presence of said agent;
- and
- comparing the level of phosphorylation of said substrate from (a) with the level of phosphorylation from (b).
