Abstract
The invention is directed to compositions of mutated binding proteins containing thiol groups for coupling to sensor surfaces, analyte biosensor devices derived there from, and methods of their use as analyte biosensors both in vitro and in vivo.
Claims22
A method for detecting the concentration of an analyte in a sample, the method comprising:
- a. exposing the sample to a biosensor comprising (i) at least one mutated glucose/galactose binding protein having at least one substituted or added cysteine group, and (ii) at least one sensor surface wherein said mutated binding protein is attached to said surface through said at least one cysteine group;
- and
- b. detecting a signal from the biosensor upon binding of the analyte to said mutated binding protein,
- wherein the signal is indicative of the concentration of the analyte in the sample.
