Abstract
Disclosed herein are methods and materials for producing a more developmentally potent cell from a less developmentally potent cell. Specifically exemplified herein are methods that comprise introducing an expressible dedifferentiating polynucleotide sequence into a less developmentally potent cell, wherein the transfected less developmentally potent cell becomes a more developmentally potent cell capable of differentiating to a less developmentally potent cell of its lineage of origin or a different lineage.
Claims12
A method of increasing potency of a cell comprising introducing into said cell a polynucleotide encoding a polypeptide exhibiting dedifferentiating influence.
A method of increasing potency of a mesenchymal stem cell comprising introducing into said cell a polynucleotide sequence encoding a nanog, or a polynucleotide sequence that hybridizes to a complement of the sequence in FIG. 9 under high stringency conditions which encodes a polypeptide sequence exhibiting dedifferentiating influence on mesenchymal stem cells.
A method of transforming an adult human mesenchymal cell into a pluripotent cell comprising (a) introducing into said cell a polynucleotide sequence that hybridizes to a complement of the sequence in FIG. 9 under high stringency conditions which encodes a peptide sequence exhibiting dedifferentiating influence on mesenchymal stem cells;
- and (b) presenting the cell from step (a) in a tissue culture environment to allow for dedifferentiation into a pluripotent state.
A method of producing an autologous dedifferentiated cell sample comprising obtaining a sample of adult mesenchymal stem cells from a subject;
- introducing into cells from said sample a polynucleotide sequence that hybridizes to a complement of the sequence in FIG. 9 under high stringency conditions which encodes a peptide sequence exhibiting dedifferentiating influence on mesenchymal stem cells, thereby forming transfected cells;
- and providing said transfected cells in a tissue culture environment to allow for dedifferentiation of said transfected cells.
