Abstract
The present invention relates to ubiquitinated TNF receptor 2, and the uses thereof. More specifically, the invention relates to the use of TNF receptor 2 ubiquitination to deplete TNF receptor 2 from the cell membrane and cytoplasm, and relocalize it in the insoluble cell fraction. Such relocalization can be used to modulate the signaling activity of the TNF receptor 2 and to treat TNF receptor 2-related diseases. The invention relates further to the use of Smurf 2 to ubiquitinate TNF receptor 2 and to the use of TRAF2 to mediate TNF receptor 2 ubiquitination.
Claims16
A tumor necrosis factor receptor 2 (TNF-R2) comprising SEQ ID NO:1 or a functional fragment thereof,
- wherein said TNF-R2 or functional fragment is ubiquitinated.
A process for producing ubiquinated tumor necrosis factor receptor 2 (TNF-R2), said process comprising:
- ubiquinating TNF-R2 in the presence of Smurf2.
A method of mediating tumor necrosis factor receptor 2 (TNF-R2) ubiquitination, said method comprising:
- ubiquinating TNF-R2 in the presence of tumor necrosis factor receptor-associated factor 2 (TRAF2) so as to mediate the TNF-R2 ubiquitination.
A method of relocalizing tumor necrosis factor receptor 2 (TNF-R2) to an insoluble cell fraction, said method comprising:
- ubiquinating TNF-R2 or using Smurf2 and/or tumor necrosis factor receptor-associated factor 2 (TRAF2),
- so as to relocalize TNF-R2 to the insoluble cell fraction.
A method of modulating tumor necrosis factor receptor 2 (TNF-R2) signaling activity, said method comprising:
- ubiquinating TNF-R2 or using Smurf2 and/or tumor necrosis factor receptor-associated factor 2 (TRAF2) and/or Smurf2 or TRAF2 modulating reagents
- so as to modulate the TNFR-2 signaling activity.
A method of modulating apoptosis in a cell, said method comprising:
- modulating apoptosis with tumor necrosis factor receptor 2 (TNF-R2) ubiquitination so as to modulate apoptosis in the cell.
A method of modulating inflammation or auto-immunity associated with a disease state in a subject, said method comprising:
- ubiquinating tumor necrosis factor receptor 2 (TNF-R2) in the subject so as to modulate inflammation or auto-immunity.
A method of modulating cell proliferation, said method comprising:
- ubiquinating tumor necrosis factor receptor 2 (TNF-R2) in a cell population
- so as to modulate cell proliferation.
