Abstract
The present invention provides a new and improved method for quantitative determination and/or identification of proteins in a sample. In one aspect, the invention provides a mass spectroscopic method for comparing protein levels in two or more samples by differentially isotopically labeling each sample's proteins' N- or C-termini. In another aspect, the invention provides a mass-spectroscopic method for identifying a sample as source for a protein from a mixture of two or more samples by differentially isotopically labeling each sample's proteins' N- or C-termini.
Claims28
A mass spectroscopic method for comparing protein levels in two or more samples comprising steps of:
- a. providing two or more samples each containing one or more proteins;
- b. differentially labeling each samples' proteins N- or C- termini with a detectable isotopic label to generate two or more sets of differentially isotopically labeled proteins;
- c. measuring the relative levels of differentially isotopically labeled proteins by a mass spectroscopic technique.
A mass-spectroscopic method for identifying a sample as source for a protein from a mixture of two or more samples, the method comprising:
- a. providing two or more samples each containing one or more proteins;
- b. differentially labeling each sample's proteins' N- or C-termini with a detectable isotopic label to generate two or more sets of differentially isotopically labeled proteins;
- c. mixing the labeled proteins from the two or more samples;
- d. analyzing the mixture of labeled proteins using a mass spectroscopy technique;
- e. detecting a fragment ion from the label portion of a labeled protein thereby identifying the proteomic sample source of the labeled protein.
