Abstract
The invention provides extraction columns for the purification of an analyte (e.g., a biological macromolecule, such as a peptide, protein or nucleic acid) from a sample solution, as well as methods for making and using such columns. The columns typically include a bed of extraction media positioned in the column, often between two frits. In some embodiments, the extraction columns employ modified pipette tips as column bodies. In some embodiments, the invention provides methods characterized by the use of hydrophobic and/or hydrophilic membranes as frits, and/or by the inclusion of a wad of fibrous material to act as a wicking agent.
Claims21
A sample processing column comprising:
- i) a column body having an open upper end, an open lower end, and an open channel between the upper and lower ends of the column body;
- ii) a bottom frit bonded to and extending across the open lower end, wherein said bottom frit is hydrophilic;
- and
- iii) a bed of extraction media positioned inside the open channel and in contact with the bottom frit, wherein said bed of extraction media is comprised of gel resin beads.
The method of claim 12, wherein between steps (ii) and (iii) a wash solution is aspirated into the bed of extraction media and dispensed from the bed of extraction media.
The purification system of claim 22, wherein the open lower end of the sample processing columns are in contact with a solution selected from the group consisting of the sample solution, the wash solution, and the desorption solution.
