Abstract
The invention relates to a recombinant DNA and polypeptide sequence of an immuno-dominant phage particle associated protein from Bartonella bacilliformis. The recombinant protein is easily produced permitting the conduct of more accurate and rapid diagnositic assays for the detection of B. bacilliformis infection with reduced reagent and equipment requirements over that required by currently available methods of diagnosis. The DNA and polypeptide sequence is also useful in vaccine preparations against B. bacilliformis.
Claims16
A nucleotide sequence encoding an immunogenic polypeptide of the Pap31 sequence of a Bartonella bacilliformis bacteriophage wherein said nucleotide sequence is SEQ ID No. 1 or a portion of SEQ ID No. 1 encoding at least one antibody epitope.
A polypeptide sequence encoding an immunogenic polypeptide of the Pap31 sequence of Bartonella bacilliformis bacteriophage wherein said polypeptide sequence is SEQ ID No. 2 or a portion of SEQ ID No. 2 encoding at least one antibody epitope.
A method for detecting B. bacilliformis infection, wherein detection is by antibody-based methods comprises the steps:
- a. attaching polypeptide or fragments of said method antibody-based assay comprising a polypeptide sequence encoded by all or part of the sequence encoded by SEQ ID No. 2 to a solid matrix;
- b. adding sample antibody to said polypeptide attached surface
- c. detecting binding of said sample antibody to said polypeptide.
A method of diagnosis according to claim 11 wherein said method comprises the additional steps of:
- d. exposing a range of known concentrations of Pap31-specific antibody positive and negative control antibody to said surface attached polypeptides as in steps (a) through (c);
- e. measuring the binding of said positive and negative control antibody to the said surface-attached polypeptides;
- f. comparing said binding of positive and negative control antibody to the binding of said sample antibody to obtain an antibody concentration estimate in said sample antibody.
An immunogenic composition against Bartonella bacilliformis encoding the polypeptide sequence of SEQ ID No. 2 or a portion of SEQ ID No 2 encoding at least one T or B cell epitope.
