Abstract
A substrate for testing a carrier liquid for biomolecules using SPR or other techniques provides a metallic island surrounded by a hydrophobic layer, the islands being a location for the attachment of probe molecules. The hydrophobic layer may also be a dielectric material providing improved sensitivity in SPR imaging.
Claims42
A substrate for use in surface plasmon resonance (SPR) comprising:
- a support surface transmitting SPR analysis light;
- a plurality of islands of metallic film attached to the support surface to expose a metallic surface providing spatially isolated regions at which probe molecules may be attached to react in situ with a target material in a carrier solution coming into contact with an exposed metallic surface;
- and
- a hydrophobic layer adhered to the support surface surrounding the plurality of islands.
A method of constructing a substrate for use in chemical analyses comprising the steps of
- (a) coating a support surface with a hydrophobic layer;
- and
- (b) attaching a plurality of islands of metallic film to the hydrophobic layer to expose a metallic surface providing spatially isolated regions in which probe molecules may be attached to react in situ with a target material in a carrier solution coming into contact with an exposed metallic surface.
A substrate for use in surface plasmon resonance imaging comprising:
- a transparent support substrate passing analyzing light therethrough;
- a dielectric layer adhered to a surface of the transparent substrate to expose a dielectric surface;
- a metallic film adhered to at least a portion of the dielectric surface to expose a metallic surface;
- and
- probe molecules attached to the metallic film to react in situ with a target material in a carrier solution coming into contact with the exposed metallic surface;
- wherein the dielectric layer is selected to promote long-range surface plasmon resonance.
A method of chemical analysis comprising the steps of
- (a) preparing a transparent support substrate having a metallic film adhered to a dielectric layer in turn attached to the support substrate;
- (b) attaching at least one probe molecule to the metallic film;
- (c) reacting probe molecules attached to the metallic film with a target material in a carrier solution coming into contact with an exposed metallic film surface;
- and
- (d) detecting the reaction using variations in reflectivity caused by surface plasmon resonance.
A method of chemical analysis comprising the steps of
- (a) preparing a transparent support substrate having a metallic film supported by the transparent substrate;
- (b) attaching probe molecules to the metallic film at different predetermined locations;
- (c) reacting the probe molecules as attached to the metallic film with at least one labeled target material in a carrier solution coming into contact with the exposed metallic film surface;
- (d) detecting the reaction using variations in reflectivity of light passing through the transparent support substrate to reflect off of the metallic film at the different locations;
- (e) detecting the reaction through sensing of labeling of the target material at the different locations.
