Abstract
Methods of increasing yields of succinate using aerobic culture methods and a multi-mutant E. coli strain are provided. Also provided is a mutant strain of E. coli that produces high amounts of succinic acid.
Claims20
A genetically engineered bacterial strain wherein said bacteria comprise:
- a) a reduced activity of acetate kinase-phosphotransacetylase (ACKA-PTA);
- and
- b) a reduced activity for one or more proteins selected from the group consisting of aerobic respiratory control regulator A and B (ARCAB), fatty acid degradation regulon (FADR), fumarate reductase (FRD), fructose regulon (FRUR), fumarase (FUMA, FUMB, or FUMC), isocitrate dehydrogenase (ICD), aceBAK operon repressor (ICLR), malate dehydrogenase (MDH), malate synthase (MS), pyruvate formate lyase (PFL), pyruvate oxidase (POXB), phosphotransferase system proteins F or G (PTSF or PTSG), and succinate dehydrogenase (SDH).
A method of producing tricarboxylic acids in an aerobic bacterial culture comprising:
- a) providing a bacteria in a culture wherein said bacteria comprises reduced activity of acetate kinase-phosphotransacetylase (ACKA-PTA),
- wherein said bacteria is selected from the group consisting of:
- i) bacteria comprising a reduced activity for one or more proteins selected from the group consisting of aerobic respiratory control regulator A and B (ARCAB), fatty acid degradation regulon (FADR), fumarate reductase (FRD), fructose regulon (FRUR), fumarase (FUMA, FUMB, or FUMC), isocitrate dehydrogenase (ICD), aceBAK operon repressor (ICLR), malate dehydrogenase (MDH), malate synthase (MS), pyruvate formate lyase (PFL), pyruvate oxidase (POXB), phosphotransferase system proteins F or G (PTSF or PTSG), and succinate dehydrogenase (SDH);
- ii) a bacteria comprising overexpression of a protein selected from the group consisting of phosphoenolpyruvate carboxylase (PEPC), isocitrate lyase (ACEA), malate synthase (ACEB) and isocitrate dehydrogenase kinase/phosphorylase (ACEK);
- and
- iii) a bacteria comprising both i) and ii);
- b) supplying the bacteria with a sugar substrate,
- c) allowing the bacteria to metabolize the sugar, and
- d) recovering the tricarboxylic acid from the culture.
