Abstract
The invention relates to the production of adult dedifferentiated, programmable stem cells from human monocytes by cultivation of monocytes in a culture medium which contains M-CSF and IL-3. The invention further relates to pharmaceutical preparations, which contain the dedifferentiated, programmable stem cells and the use of these stem cells for the production of target cells and target tissue.

1-34. (canceled)
A method for forming dedifferentiated programmable stem cells derived from human monocytes, which comprises:
- providing a monocyte;
- and adding M-CSF and IL-3 to form a dedifferentiated programmable stem cell derived from said monocyte.
A method for making dedifferentiated stem cells of monocytic origin, which comprises:
- providing an in vitro culture of monocytes;
- and adding macrophage colony stimulating factor (M-CSF) in a concentration of 2-20 nanogram/mL (ng/ml) and interleukin 3 (IL-3) in a concentration of 0.2-1 ng/ml.
A method for making conditioned medium, which comprises:
- providing an in vitro culture of monocytes after treatment with M-CSF and IL-3 from day 0 to the day 6;
- and subsequently treating the culture with a differentiation medium.
A cultured cell from a dedifferentiated, programmable stem cell derived from a monocyte, comprising a protein, said protein being selected from the group consisting of a cell-surface-membrane protein.
A dedifferentiated, programmable stem cell derived from a monocyte for differentiation into many different cell types, developing function and morphology of nearly all other human cells.
A method for differentiating a dedifferentiated, programmable stem cell derived from a monocyte, which comprises:
- maintaining a culture media through new cell cycles from day 0 to day 6;
- and differentiating said dedifferentiated, programmable stem cell derived from a monocyte by supplementing said dedifferentiated, programmable stem cell derived from a monocyte with a conditioned medium between day 0 to 6 into a differentiated, human, specific cell.
A method for detecting dedifferentiated, programmable stem cells of monocytic origin incubated in pancreatic extract, which comprises detecting a pancreatic protein with corresponding specific antibodies.
The method according to claim 32, wherein the pancreatic protein is insulin.
A method for detecting dedifferentiated, programmable stem cells of monocytic origin incubated with a mixture of liver cells in culture medium, which comprises detecting a liver protein with specific monoclonal antibodies by immune-histochemistry.
The method according to claim 34, wherein the liver protein is albumin.
A method for treating tissue, which comprises applying in vivo a dedifferentiated, programmable stem cell of monocytic origin to the tissue.
A method for in vitro induction of pluripotency including the corresponding immunophenotype in human adherent mononuclear cells rich in monocytes with the immunophenotype detailed above.
