Abstract
The present invention relates to isolated and purified Magmas protein, isolated and purified nucleic acids encoding Magmas protein, and antibodies to Magmas protein, which interacts with granulocyte macrophage colony stimulating factor (GM-CSF). More specifically, the invention relates to uses of such anti-Magmas antibodies. The invention further relates to a method for diagnosis, prognosis and treatment of diseases, particularly, cancer, Alzheimer's disease and mitochondrial diseases using Magmas sequences and antibodies directed against the Magmas protein or fragments thereof.
Claims25
An antibody that specifically binds to a Magmas protein, wherein said protein comprises SEQ ID NO:
- 4 or has at least 85% homology with SEQ ID NO:
- 4, as defined using BLAST at default settings.
An isolated nucleic acid sequence comprising SEQ ID NO:
- 11 or a fragment of at least 20 contiguous nucleotides thereof.
An isolated nucleic acid sequence encoding SEQ ID NO:
- 4 or a fragment of at least 6 contiguous amino acids thereof.
A method of identifying a disease condition comprising obtaining a biological sample from an individual and analyzing either the nucleic acid of SEQ ID NO 11 or the protein of SEQ ID NO:
- 4 in the sample, wherein a variation in SEQ ID NO:
- 11 or SEQ ID NO:
- 4 indicates a disease condition.
A method of detecting cancer comprising the steps of:
- a) providing a biological sample from an individual suspected of having cancer;
- b) analyzing the Magmas protein expression in the sample;
- and
- c) comparing the expression of Magmas protein in the sample against a normal control, wherein increased expression of the Magmas protein in the sample indicates that the individual is at increased risk for cancer.
A method of determining the severity of prostate cancer comprising the steps of:
- a) taking a biological sample of an individual suspected of having prostate cancer;
- b) analyzing the Magmas protein expression in the sample;
- c) comparing the amount of expression of the Magmas protein in the sample to amount of expression in a panel of samples comprising of tissue samples representing different stages of prostate cancer wherein the different stages of prostate cancer are differentiated from each other by different amount of Magmas expression, wherein the expression of the Magmas protein in the sample indicates the stage of prostate cancer.
A method of diagnosing Alzheimer's disease in an individual comprising measuring Magmas expression in the pyramidal neurons of the hippocampus wherein reduction in the Magmas expression compared to a control indicates that the individual is affected with Alzheimer's disease.
A method of treating an individual affected with cancer comprising the steps of administering a therapeutic Magmas blocking amount of a compound that interacts with Magmas such as a Magmas antibody in a pharmaceutically acceptable carrier to an individual having cancer.
An antibody against a C-terminal fragment of SEQ ID NO:
- 4, wherein said fragment begins at amino acid 29 or higher of SEQ ID NO:
- 4.
An antibody against an N-terminal fragment of SEQ ID NO:
- 4 before the amino acid 29 of the SEQ ID NO:
- 4.
An antibody against a fragment beginning at amino acid 29 of SEQ ID NO:
- 4 or higher.
A vector comprising a nucleic acid encoding an amino acid sequence selected from the group of SEQ ID NO:
- 3, SEQ ID NO:
- 4 and SEQ ID NO:
- 11.
An isolated a purified nucleic acid encoding an amino acid sequence selected from the group consisting of SEQ ID NO:
- 5, SEQ ID NO:
- 6, SEQ ID NO:
- 7, SEQ ID NO:
- 8 and SEQ ID NO:
- 9.
A vector comprising a nucleic acid encoding an amino acid sequence selected from the group consisting of SEQ ID NO:
- 5, SEQ ID NO:
- 6, SEQ ID NO:
- 7, SEQ ID NO:
- 8 and SEQ ID NO:
- 9.
