Abstract
Diagnostic devices and methods are provided. The diagnostic devices preferably comprise a test strip with a test area. The sample to be analyzed contacts the test area, which comprises a specific binding partner for the analyte of interest. Analyte, if present in the sample, binds to the immobilized binding partner in the test area and is subsequently contacted with a conjugate. The conjugate specifically binds the analyte and provides a visual indication of the presence of the analyte. The devices may be used for the diagnosis of particular diseases or disorders in a patient, such as HIV or hepatitis. They may also be used to determine if an individual is pregnant.
Claims44
A diagnostic test strip for detecting an analyte of interest in a liquid sample, the test strip comprising:
- a conjugate source area, comprising a conjugate configured to bind the analyte of interest;
- a sample application area located downstream of said conjugate source area and configured to receive said liquid sample;
- and
- a test area located downstream of said conjugate source area and comprising immobilized binding agent specific for the analyte of interest.
A test strip for the detection of anti-HIV antibody in a sample comprising:
- a test area and a conjugate source area located upstream of said test area;
- immobilized antigen bound at said test area, wherein the immobilized antigen comprises about 0.1 to about 0.5 μg of HIV antigen;
- and
- a conjugate diffusably bound to said conjugate source area, wherein the conjugate is configured to bind an antibody.
A method of detecting an antibody of interest in a liquid sample comprising:
- contacting the test area of a of a test strip with the sample, wherein the test area comprises immobilized antigen that is specific for the antibody of interest;
- and
- subsequently contacting the test area with a conjugate configured to bind the antibody of interest.
An immunodiagnostic pregnancy test strip comprising:
- a sample application area for receiving a liquid sample;
- a test area comprising an immobilized first anti-hCG antibody;
- and
- a conjugate source area comprising conjugate configured to bind hCG,
- wherein said conjugate source area is located upstream of said sample application area and said test area.
