Abstract
The preparation and characterization of antibodies that bind to antigens on CLL or other cancer cells, especially to antigens upregulated in the cancer cells, and the identification and characterization of antigens present on or upregulated by cancer cells are useful in studying and treating cancer.
Claims28
A method for identifying an antigen to which an antibody binds comprising:
- preparing a library of antibodies using tissue harvested from a CLL patient;
- contacting one or more members of the library with a lysate of the cell line CLL-MT, deposited under ATCC Accession No. PTA-3920;
- and
- characterizing an antigen bound to at least one member of the library.
An antibody that binds to an antigen that is upregulated by a CLL cell.
A method for identifying an antigen to which an antibody binds comprising:
- preparing a library of antibodies using cells in the cell line CLL-AAT, deposited under ATCC Accession No. PTA-3920;
- contacting one or more members of the library with a lysate of cells from tissue harvested from a CLL patient;
- and
- characterizing an antigen bound to at least one member of the library.
A method for identifying an antigen to which an antibody binds comprising:
- immunizing an organism with at least a portion of a CLL cell from tissue harvested from a CLL patient;
- generating an antibody library based on the immune response of the organism to immunization;
- contacting one or more members of the library with CLL cells;
- and
- characterizing an antigen bound to at least one member of the library.
A method for identifying an antigen to which an antibody binds comprising:
- immunizing an organism with at least a portion of a CLL cell;
- generating an antibody library based on the immune response of the organism to immunization;
- contacting one or more members of the library with a lysate of the cell line CLL-AAT, deposited under ATCC Accession No. PTA-3920 one or more members of the library with CLL cells;
- and
- characterizing an antigen bound to at least one member of the library.
A method of treating cancer comprising administering an antibody that interferes with the metabolic pathway of a polypeptide that is upregulated by a malignant cancer cell.
A method comprising:
- screening cancer patients to identify those in which expression of a polypeptide is upregulated by a malignant cancer cell;
- and
- administering an antibody that interferes with the metabolic pathway of the upregulated polypeptide to those patients in which upregulation is found.
