Abstract
A means and method for treating bacterial infection, providing antioxidant activity, and chelating copper using a copper binding compound produced by methanotrophic bacteria is described. The compound, known as methanobactin, is the first of a new class of antibiotics having gram-positive activity. Methanobactin has been sequenced, and its structural formula determined.
Claims36
A copper binding protein having the sequence N-2-isopropyloxycarbonyl-4-hydroxy-5-thionyl-imidazole-Ser-Cys-Tyr-pyrrolidine-4-hydroxy-5-thiony-imidazole-Ser-Cys-Met.
A pharmaceutical composition comprising:
- methanobactin;
- and a pharmaceutically acceptable carrier.
A method of preventing or treating a bacterial infection comprising:
- administrating a bacterial infection preventative or bacterial treatment effective amount of methanobactin.
A method of providing anti-oxidant activity to an animal comprising:
- administrating to the animal an anti-oxidant effective amount of methanobactin.
A method of chelating copper comprising:
- administering a copper-chelating effective amount of methanobactin.
A method of preparing a methanobactin pharmaceutical composition comprising:
- culturing a methanotroph under conditions whereby the methanotroph is deprived of copper to produce methanobactin;
- separating the methanotrophs from the methanobactin;
- and combining the methanobactin with a pharmaceutically acceptable carrier.
