Abstract
The present invention is directed to a composition and method for enhancing the complement-mediated cytotoxicity of therapeutic antibodies for their target cells (i.e. those cells displaying the target epitope). More particularly the present invention enhances the efficacy of such therapies by providing a supplemental source of complement
Claims26
A method of enhancing the complement mediated cytotoxic activity of an anti-neoplastic antibody for its target cells, said method comprising the steps of
- contacting said cells with the anti-neoplastic antibody;
- and
- contacting the cells with a composition comprising a donor complement protein.
A kit for enhancing the cytotoxic activity of anti-tumor antibodies, said kit comprising a composition wherein the composition comprises a complement protein.
A composition for treating patients with a neoplastic disease, said composition comprising
- an anti-neoplastic antibody;
- and
- a complement protein.
A method for identifying individuals that would benefit from supplementing an anti-cancer antibody therapy with donor complement, said method comprising the steps of:
- providing a serum or plasma sample from the individual to be treated;
- contacting the sample with cancer cells and the anti-cancer antibody to form a reaction mixture;
- incubating the reaction mixture in the presence and absence of donor complement proteins;
- and
- determining whether the sample exhibits higher opsonization or cell killing in the presence of supplement complement as the criteria for identifying a individual that would benefit from complement supplementation.
