Abstract
The invention relates a nucleic acid and its encoded polypeptide ESM-1, whose expression is modulated in angiogenesis and oncogenesis. The invention also relates to antibodies having specificity for said polypeptide. The present invention also relates to antisense molecules. The invention further relates to methods useful for treating or modulating angiogenesis in mammals in need of such biological effect.
Claims41
An isolated ESM-1 polypeptide comprising an amino acid sequence selected from the group consisting of:
- (a) an amino acid sequence comprising a sequence of SEQ ID NO:2;
- (b) a variant of an amino acid sequence comprising a sequence of SEQ ID NO:2, wherein one or more amino acid residues in said variant differs from the amino acid sequence of said mature form, provided that said variant differs in no more than about 30% of amino acid residues from said amino acid sequence;
- (c) a secreted mature form of an amino acid sequence of SEQ ID NO:19;
- (d) a variant of a mature form of an amino acid sequence of SEQ ID NO:19, wherein one or more amino acid residues in said variant differs from the amino acid sequence of said mature form, provided that said variant differs in no more than about 30%, of the amino acid residues from the amino acid sequence of said mature form;
- and
- (e) a fragment of the amino acid sequence of SEQ ID NO2, or SEQ ID NO:19.
An isolated ESM-1 polypeptide comprising an amino acid sequence selected from the group consisting of:
- SEQ ID NO:2, or SEQ ID NO:3.
A method of detecting differentially expressed genes correlated with a cancerous state of a mammalian cell, the method comprising the step of detecting at least one differentially expressed gene product in a test sample derived from a cell suspected of being cancerous, where the gene product is encoded by the nucleic acid sequence SEQ ID NO:1, wherein detection of differentially expressed product is correlated with a cancerous state of the cell from which the test sample was derived.
A method of selecting a composition for inhibiting angiogenesis in a patient, the method comprising:
- (a) obtaining a sample comprising cancer cells from the patient;
- (b) separately exposing aliquots of the sample in the presence of a plurality of test compositions;
- (c) comparing expression of a marker in each of the aliquots, wherein the marker is selected from the group consisting of the markers of SEQ ID NO:2, and SEQ ID NO:3, and
- (d) selecting one of the test compositions which alters the level of expression of the marker in the aliquot containing that test composition, relative to other test compositions.
An ESM-1 polypeptide antagonist.
A transgenic knockout non-human mammal comprising a homozygous disruption in its endogenous ESM-1 gene, wherein said disruption prevents the expression of a functional ESM-1 protein.
