Abstract
The invention provides a self-cleaving protein or intein. The intein can be obtained from a wide range of Cryptococcus neoformans. Also provided are methods of using the inteins such as in protein purification and as a target in testing the efficacy of drugs to inhibit intein function.

An isolated intein obtainable from Cryptococcus neoformans or a functionally equivalent, or functionally altered, fragment or variant thereof.
An intein having the amino acid sequence of SEQ ID NO:1.
An isolated intein which has an amino acid sequence which has greater than about 35% identity with the sequence of SEQ ID NO:1.
An isolated nucleic acid molecule which encodes an intein which is part of the genome of C. neoformans strain Cn 3511, on deposit at American Type Culture Collection, Maryland, USA, under accession number ATCC 32045.
