Abstract
1 Compounds of formula (I) are antibacterial or antiprotozoal agents for treatment of infections in humans and non-human mammals, wherein: Z represents a radical of formula —N(OH)CH(═O) or formula C(═O)NH(OH); R1 represents hydrogen, methyl or trifluromethyl, or except when Z is a radical of formula —N(OH)(═O), a hydroxy, halo or am group; R2 represents a group R10—(X)n-(ALK)m— wherein R10, ALK, X, m and n are as defined in the specification; R3 represents hydrogen, (C1-C6) alkyl or phenyl(C1-C6)alkyl-; and R5 and R6 are as defined in the specification.
Claims16
The use of a compound of formula (I) or a pharmaceutically or veterinarily acceptable salt, hydrate or solvate thereof in the preparation of a composition for treatment of bacterial or protozoal infections in humans and non-human mammals:
- 22
- wherein:
- Z represents a radical of formula —N(OH)CH(═O) or formula —C(═O)NH(OH);
- R1 represents hydrogen, methyl or trifluoromethyl, or, except when Z is a radical of formula —N(OH)CH(═O), a hydroxy, halo or amino group;
- R2 represents a group R10—(X)n-(ALK)m— wherein
- R10 represents hydrogen, or a C1-C6 alkyl, C2-C6 alkenyl, C2-C6 alkynyl, cycloalkyl, aryl, or heterocyclyl group, any of which may be unsubstituted or substituted by (C1-C6)alkyl, (C1-C6)alkoxy, hydroxy, mercapto, (C1-C6)alkylthio, amino, halo (including fluoro, chloro, bromo and iodo), trifluoromethyl, cyano, nitro, oxo, —COOH, —CONH2, —COORA, —NHCORA, —CONHRA, —NHRA, —NRARB, or —CONRARB wherein RA and RB are independently a (C1-C6)alkyl group and
- ALK represents a straight or branched divalent C1-C6 alkylene, C2-C6 alkenylene, or C2-C6 alkynylene radical, and may be interrupted by one or more non-adjacent —NH—, —O— or —S— linkages,
- X represents —NH—, —O— or —S—, and
- m and n are independently 0 or 1;
- R3 represents the side chain of a natural or non-natural alpha amino acid;
- and
- R4 represents hydrogen, (C1-C6)alkyl or phenyl(C1-C6)alkyl-;
- and
- (a) R5 and R6 independently represent:
- hydrogen, halogen, trifluoromethyl, or
- (C1-C6)alkyl, (C2-C6)alkenyl, (C2-C6)alkynyl, aryl, heterocyclyl, aryl(C1-C6)alkyl-, or heterocyclyl(C1-C6)alkyl-, any of which may be optionally substituted by
- (C1-C6)alkyl, halogen, trifluoromethyl, cyano, nitro, oxo,
- phenoxy or phenylthio-,
- —ORA, —SRA, —NHRA, —NRARB, —NHCORA—CONHRA, —CONRARB, or —COORA, wherein
- RA and RB are independently hydrogen or (C1-C4)alkyl,
- or in the case where RA and RB are attached to a nitrogen atom then RA and RB taken together with the nitrogen atom to which they are attached form a monocyclic 5-7 membered ring;
- or
- (b) R5 and R6 taken together with the carbon atoms to which they are attached form a fused monocyclic or bicyclic carbocyclic or heterocyclic ring which may be further substituted with any of the substituent groups listed under (a).
A compound as claimed in claim 14 wherein R1 is hydrogen, R2 is n-propyl, n-butyl, n-pentyl, or cyclopentylmethyl, R3 is tert-butyl, and R4 is hydrogen or methyl.
