Abstract
The invention provides isolated Bcl-2 domain-containing polypeptides from Mycobacterial species, including M. tuberculosis, M. avium, M. bovis, M. leprae and M. smegmatis, and from Streptomyces species, including S. coelicolor, as well as modifications of such polypeptides, functional fragments therefrom, encoding nucleic acid molecules and specific antibodies. Also provided are methods for identifying polypeptides and compounds that associate with or modulate the activity of the Bcl-2 domain-containing polypeptides. Further provided are methods of modulating apoptosis and treating pathological conditions using the described nucleic acid molecules, polypeptides and compounds.
Claims34
An isolated nucleic acid molecule encoding a polypeptide comprising a Bcl-2 domain, selected from the group consisting of:
- (a) a nucleic acid molecule encoding a polypeptide comprising the amino acid sequence designated SEQ ID NOS:5, 9, 13, 17, 21 or 25;
- and
- (b) a nucleic acid molecule that hybridizes to the nucleic acid molecule of (a) or its complement under highly stringent hybridization conditions.
An isolated oligonucleotide comprising at least 17 contiguous nucleotides of the nucleotide sequence set forth as SEQ ID NOS:4, 8, 12, 16, 20 or 24, or the complement thereof.
An isolated polypeptide comprising a Bcl-2 domain, wherein said polypeptide comprises the amino acid sequence designated SEQ ID NOS:5, 9, 13, 17, 21 or 25, or modification thereof comprising an amino acid sequence at least 80% identical thereto.
An isolated peptide comprising at least 8 contiguous amino acids of the amino acid sequence set forth as SEQ ID NOS:5, 9, 13, 17, 21 or 25.
A method of modulating apoptosis in a cell, comprising introducing into said cell an antisense, dsRNA or ribozyme nucleic acid molecule, wherein said nucleic acid molecule specifically hybridizes to SEQ ID NOS:1, 6, 10, 14, 18 or 22.
A method of treating a pathological condition in an individual, comprising introducing into a diseased cell in said individual an antisense, dsRNA or ribozyme nucleic acid molecule, wherein said nucleic acid molecule specifically hybridizes to SEQ ID NOS:1, 6, 10, 14, 18 or 22.
