An influenza viral vector, comprising influenza virus incorporation sequences, which vector comprises:
- a) sequences corresponding to the 3′ noncoding region of influenza virus HA vRNA, 3′ NA incorporation sequences, a heterologous nucleic acid segment, and the 5′ noncoding region of NA vRNA;
- b) sequences corresponding to the 3′ noncoding region of influenza virus HA vRNA, a heterologous nucleic acid segment, 5′ HA incorporation sequences, and the 5′ noncoding region of HA vRNA;
- c) sequences corresponding to the 3′ noncoding region of influenza virus M vRNA, 3′ M incorporation sequences, a heterologous nucleic acid segment, 5′ M incorporation sequences, and the 5′ noncoding region of M vRNA;
- d) sequences corresponding to the 3′ noncoding region of influenza virus NS vRNA, 3′ NS incorporation sequences, a heterologous nucleic acid segment, and the 5′ noncoding region of NS vRNA;
- e) sequences corresponding to the 3′ noncoding region of influenza virus PB2 vRNA, a heterologous nucleic acid segment, 5′ PB2 incorporation sequences, and the 5′ noncoding region of PB2 vRNA;
- f) sequences corresponding to the 3′ noncoding region of influenza virus PB1 vRNA, a heterologous nucleic acid segment, 5′ PB1 incorporation sequences, and the 5′ noncoding region of PB1 vRNA;
- or
- g) sequences corresponding to the 3′ noncoding region of influenza virus PA vRNA, a heterologous nucleic acid segment, 5′ PA incorporation sequences, and the 5′ noncoding region of PA vRNA;
- wherein vRNA corresponding to the vector, when present in a cell expressing influenza virus proteins and comprising vRNAs other than the vRNA corresponding to the vector, is packaged into virions.
An isolated nucleic acid molecule comprising influenza virus NA incorporation sequences corresponding to the 5′ coding region of NA and a heterologous nucleic acid segment.
An isolated nucleic acid molecule comprising influenza virus NS incorporation sequences corresponding to the 5′ coding region of NS and a heterologous nucleic acid segment.
An isolated nucleic acid molecule comprising influenza virus HA incorporation sequences corresponding to the 3′ coding region of HA and a heterologous nucleic acid segment.
An isolated nucleic acid molecule comprising influenza virus PB2 incorporation sequences corresponding to the 3′ coding region of PB2 and a heterologous nucleic acid segment.
An isolated nucleic acid molecule comprising influenza virus M incorporation sequences corresponding to the 3′ coding region of M and the 5′ coding region of M, and a heterologous nucleic acid segment.
A method to prepare replication incompetent influenza-like virus, comprising:
- a) contacting a recombinant host cell with a recombinant influenza virus which lacks a vRNA with a wild-type NS coding sequence and which comprises sequences for a recombinase, wherein the recombinant host cell comprises a recombinant nucleic acid molecule comprising a NS2 open reading frame but not an incorporation sequence operably linked to two site-specific recombination sequences recognized by the recombinase 5′ to the NS2 open reading frame, and wherein in the absence of the recombinase, NS2 is not expressed;
- and
- b) isolating replication incompetent influenza-like virus from the recombinant host cell.
