Abstract
Disclosed are genes found in Streptococcus pneumoniae that are essential for survival for a wide range of bacteria. These genes are termed “S-ylqF,” “S-yqeG,” “S-yybQ,” “S-yerL,” and “S-ysxC.” These genes and the polypeptides that they encode, as well as homologs and orthologs thereof can be used to identify antibacterial agents for treating a broad spectrum of bacterial infections.
Claims29
A method for identifying an antibacterial agent, the method comprising:
- (a) contacting a test compound with a polypeptide selected from the group consisting of S-ylqF, S-yqeG, S-yybQ, S-yerL, S-ysxC, B-ylqF, B-yqeG, B-yybQ, B-yerL, and B-ysxC;
- and
- (b) detecting an interaction of the test compound with the polypeptide, wherein an interaction indicates that the test compound is an antibacterial agent.
A method for identifying an antibacterial agent, the method comprising:
- (a) contacting a test compound with a polypeptide selected from the group consisting of S-ylqF, S-yqeG, S-yybQ, S-yerL, S-ysxC, B-ylqF, B-yqeG, B-yybQ, B-yerL, and B-ysxC;
- (b) detecting a decrease in function of the polypeptide contacted with the test compound;
- and
- (c) determining whether the test compound inhibits growth of bacteria, relative to growth of bacteria cultured in the absence of the test compound, wherein inhibition of growth indicates that the test compound is an antibacterial agent.
A method for identifying an antibacterial agent, the method comprising:
- (a) contacting a test compound with a nucleic acid encoding a polypeptide selected from the group consisting of S-ylqF, S-yqeG, S-yybQ, S-yerL, S-ysxC, B-ylqF, B-yqeG, B-yybQ, B-yerL, and B-ysxC;
- and
- (b) detecting an interaction of the test compound with the nucleic acid, wherein an interaction indicates that the test compound is an antibacterial agent.
A method for identifying an antibacterial agent, the method comprising:
- (a) contacting a test compound with an ortholog of a polypeptide selected from the group consisting of S-ylqF, S-yqeG, S-yybQ, S-yerL, and S-ysxC;
- and
- (b) detecting an interaction of the test compound with the ortholog, wherein an interaction indicates that the test compound is an antibacterial agent.
A method for identifying an antibacterial agent, the method comprising:
- (a) contacting a test compound with an ortholog of a polypeptide selected from the group consisting of S-ylqF, S-yqeG, S-yybQ, S-yerL, and S-ysxC;
- (b) detecting a decrease in function of the ortholog contacted by the test compound;
- and
- (c) determining whether the test compound inhibits growth of bacteria, relative to growth of bacteria cultured in the absence of the test compound, wherein inhibition of growth indicates that the test compound is an antibacterial agent.
A method for identifying an antibacterial agent, the method comprising:
- (a) contacting a test compound with a nucleic acid encoding an ortholog of a polypeptide selected from the group consisting of S-ylqF, S-yqeG, S-yybQ, S-yerL, and S-ysxC;
- and
- (b) detecting interaction of the test compound with the nucleic acid, wherein interaction indicates that the test compound is an antibacterial agent.
A method for treating a mammal having a Streptococcus pneumoniae infection, the method comprising inhibiting the function of a polypeptide selected from the group consisting of S-ylqF, S-yqeG, S-yybQ, S-yerL, and S-ysxC in Streptococcus pneumoniae infecting the mammal.
